目的 寻找mPEG BTC修饰淋巴细胞表面HLA A2抗原的最适温度 ,及如何在 37℃条件下保持修饰物的稳定。方法 在不同温度条件下 ,用终浓度为 12mmol/L的mPEG BTC在pH7.4的PBS中对淋巴细胞表面HLA A2抗原进行修饰。结果 4~ 2 5℃mPEG BTC对淋巴细胞有良好的修饰效果 ,可完全阻断淋巴细胞表面HLA A2抗原与其相应抗体的反应 ;在 30℃及 37℃条件下不利于修饰 ;在 2 2℃条件下经mPEG BTC修饰后的淋巴细胞在 37℃条件下72h内可以完全阻断HLA A2抗原与其抗体的反应。结论 在室温下经mPEG BTC修饰的淋巴细胞可以在 37℃中保持稳定。
OBJECTIVE: To find out the optimum temperature at which mPEG BTC modifies HLA A2 antigens on lymphocytes and how to maintain the stability of modification at 37 ℃. Methods The HLA A2 antigen on lymphocytes was modified with mPEG BTC at a concentration of 12 mmol / L in PBS at pH 7.4 under different temperature conditions. RESULTS: 4 ~ 25 ℃ mPEG BTC had a good modification effect on lymphocytes and completely blocked the reaction of HLA A2 antigen on the lymphocytes with its corresponding antibody. It was detrimental to modification at 30 ℃ and 37 ℃. Under the condition of 2 ℃ Under the mPEG BTC modified lymphocytes can completely block the reaction of HLA A2 antigen and its antibody within 72 hours at 37 ℃. Conclusion Lymphocytes modified with mPEG BTC at room temperature can be stable at 37 ° C.
