目的研究重组人IL-2对感染弓形虫孕鼠T细胞亚群及垂直传播的影响。方法取BALB/C孕鼠90只,随机分为空白组、感染组、IL-2组。于妊娠第8d空白对照组腹腔注射0.2ml生理盐水,其余各组腹腔注射200个弓形虫速殖子,并于妊娠第7、8、9dIL-2组每只孕鼠腹腔注射1 000UIL-2,其余两组腹腔注射0.2ml生理盐水。妊娠第10、12、14、16d取妊娠鼠脾,流式细胞仪检测CD4~+及CD8~+水平,并在妊娠第12d解剖孕鼠,观察活胎率及胎仔脑组织弓形虫感染情况。结果与空白组相比,妊娠各期感染组的脾CD4~+T细胞水平显著降低,而CD8~+T细胞水平显著升高(第10、12、1 4d),二者比值明显倒置;IL-2组的CD4~+T细胞水平在妊娠第10、12、14d高于感染组,CD8~+T细胞水平在妊娠第10、12、14d低于感染组,CD4~+/CD8~+T细胞比值在妊娠第10、12、14d高于感染组。活胎率也明显升高,宫内胚胎弓形虫感染率明显降低。结论 IL-2可以改善感染弓形虫孕鼠的细胞免疫功能,提高孕鼠免疫力,减少垂直传播概率。
Objective To study the effect of recombinant human IL-2 on T cell subsets and vertical transmission in pregnant mice infected with Toxoplasma gondii. Methods 90 BALB / C pregnant mice were randomly divided into blank group, infection group and IL-2 group. On the 8th day of gestation, the blank control group was intraperitoneally injected with 0.2ml saline, and the other groups were injected intraperitoneally with 200 Toxoplasma gondii tachyzoites. Each pregnant rat was injected intraperitoneally with 1 000UIL-2 on the 7th, The remaining two groups were injected intraperitoneally with 0.2 ml saline. Pregnant mice were sacrificed on the 10th, 12th, 14th and 16th day of gestation. The levels of CD4 ~ + and CD8 ~ + were detected by flow cytometry. Pregnant mice were dissected on the 12th day of gestation, and the live fetus rate and the Toxoplasma gondii infection were observed. Results Compared with the blank group, the levels of splenic CD4 ~ + T cells were significantly decreased and the levels of CD8 ~ + T cells were significantly increased in the infected groups (10, 12 and 14 days) The level of CD4 ~ + T cells in the -2 group was higher than that in the infection group on the 10th, 12th and 14th day of pregnancy, and the level of CD8 ~ + T cells was lower than the infected group on the 10th, 12th and 14th day of pregnancy The cell ratio was higher than the infected group on the 10th, 12th and 14th day of pregnancy. The live fetus rate was also significantly increased, Toxoplasma gondii infection rate was significantly lower. Conclusion IL-2 can improve the cellular immune function of pregnant mice infected with Toxoplasma gondii, improve the immunity of pregnant rats and reduce the probability of vertical transmission.
