目的:分析探讨塔斯品碱衍生物对人肝癌SMMC7721细胞的生长抑制作用,研究其作用机制。方法:采用MTT法测定5种塔斯品碱衍生物对肝癌SMMC7721细胞生长的抑制作用,流式细胞术检测Tas-D1对肝癌SMMC7721细胞周期的影响,ELISA法测定Tas-D1对肝癌SMMC7721细胞EGF和VEGF蛋白的影响,PCR法测定Tas-D1对EGF和VEGF的mRNA水平的影响。结果:塔斯品碱衍生物Tas-D1对SMMC7721细胞生长具有明显的抑制作用,使细胞阻滞在S期,呈现出良好的剂量依赖性。ELISA法和RT-PCR法结果表明Tas-D1对SMMC7721细胞的EGF和VEGF蛋白质的表达和mRNA的表达有一定的抑制作用(P<0.05)。结论:塔斯品碱衍生物Tas-D1能抑制人肝癌SMMC7721细胞的生长,转换细胞周期,初步作用机制主要为Tas-D1抑制SMMC7721细胞EGF和VEGF的表达。
OBJECTIVE: To investigate the inhibitory effect of taspine derivatives on the growth of human hepatocellular carcinoma SMMC7721 cells and to study its mechanism. Methods: MTT assay was used to determine the inhibitory effect of 5 taspinine derivatives on the growth of SMMC7721 cells. The effect of Tas-D1 on the cell cycle of SMMC7721 cells was detected by flow cytometry. The effect of Tas-D1 on EGF And VEGF protein. The effect of Tas-D1 on EGF and VEGF mRNA levels was determined by PCR. RESULTS: Tas-D1, a Tasasaline derivative, significantly inhibited the growth of SMMC7721 cells and blocked the cells in S phase, showing a good dose-dependent manner. The results of ELISA and RT-PCR showed that Tas-D1 could inhibit the expression of EGF and VEGF protein and the expression of mRNA in SMMC7721 cells (P <0.05). It is concluded that Tas-D1, a Tasaspin derivative, can inhibit the growth of human hepatocellular carcinoma SMMC7721 cells and change the cell cycle. The primary mechanism is that Tas-D1 inhibits the expression of EGF and VEGF in SMMC7721 cells.
