为探索更理想的过继免疫治疗手段,对由健康人外周血分离、经 CD_3单克隆抗体与白细胞介素-2共同诱导激活的 CD_3AK 细胞的体外扩增情况及其杀伤喉癌细胞 Hep-2的活性水平进行了观察和研究,并与 LAK 细胞作了比较。研究发现,CD_3AK 细胞除具有体外扩增能力强、存活时间长、IL-2依赖性低等特点外,其体内、外抗喉癌作用亦十分显著。培养早期,CD_3AK 细胞的抗瘤活性与常规培养的 LAK 细胞相似,延长培养时间至7~10天,则抗瘤活性大增,培养9天的CD_3AK 细胞在效靶比30:1的条件下平均抑瘤率高达99.8%,几乎完全抑制了喉癌细胞在裸鼠皮下的生长。研究提示,CD_3AK 细胞有望成为继 LAK 细胞之后的新一代肿瘤免疫杀伤细胞而应用于头颈肿瘤局部过继免疫治疗中。
In order to explore a more ideal means of adoptive immunotherapy, the in vitro expansion of CD_3AK cells induced by CD_3 monoclonal antibody and interleukin-2 activated by peripheral blood of healthy people and its effect on killing laryngeal carcinoma cell Hep-2 Activity levels were observed and studied and compared with LAK cells. The study found that, CD_3AK cells in addition to its ability to expand in vitro, long survival time, low IL-2-dependent characteristics, the role of anti-laryngeal cancer in vivo and in vitro is also very significant. In the early stage of culture, the anti-tumor activity of CD_3AK cells was similar to that of LAK cells in conventional culture. Prolonging the culture time to 7-10 days, the anti-tumor activity was greatly increased. CD_3AK cells cultured for 9 days averaged at the effective target ratio of 30: 1 The tumor inhibition rate was as high as 99.8%, almost completely inhibiting the growth of laryngeal carcinoma cells in nude mice. Studies suggest that CD_3AK cells are expected to become a new generation of tumor killer cells after LAK cells and be applied to the local adoptive immunotherapy of head and neck cancer.
