β-1,3-葡聚糖酶是普遍存在于多种生物中的一类多基因家族蛋白,属于PR-2家族蛋白,与植物的生长发育及防御体系有关。克隆和鉴定大豆β-1,3-葡聚糖酶家族基因对大豆抗逆育种具有重要意义。本研究应用生物信息学的方法共筛选获得45个β-1,3-葡聚糖酶基因。结果表明,GmBG1具有典型的β-1,3-葡聚糖酶家族的保守结构域及氨基酸位点,含有347个氨基酸,pI值为8.71,二级结构以ɑ螺旋为主,且N端信号肽为α螺旋结构,属于碱性蛋白。大豆β-1,3-葡聚糖酶家族基因主要在根和花中表达;实时荧光定量PCR分析表明,在腐霉菌P.ultimum的侵染下GmBG1基因在不同的抗病品种中的表达量存在差异;GmBG1基因在抗病品种中24 h时表达水平最高,而感病品种的该表达量无明显变化;此外抗病品种的该表达量明显高于感病品种。
β-1, 3-glucanase is a kind of polygene family protein which is ubiquitous in many kinds of organisms. It belongs to PR-2 family protein and is related to plant growth and defense system. Cloning and identification of soybean β-1, 3-glucanase family gene is of great significance to soybean resistance breeding. In this study, 45 β-1,3-glucanase genes were screened by bioinformatics methods. The results showed that GmBG1 has a conserved domain and amino acid sequence of typical β-1,3-glucanase family, containing 347 amino acids, with a pI value of 8.71. The secondary structure of GmBG1 is mainly helical, and the N-terminal signal Peptides are alpha helical structures and belong to basic proteins. The β-1, 3-glucanase gene family of soybean was mainly expressed in roots and flowers. Real-time quantitative PCR analysis showed that the expression level of GmBG1 gene in different resistant varieties under P. multiplication of Pythium There was a difference. The expression level of GmBG1 gene was the highest in resistant varieties at 24 h, but there was no significant change in susceptible varieties. In addition, the expression level of GmBG1 was significantly higher than that of susceptible varieties.
