观察骨质疏松大鼠不同时期骨密度 (BMD)、血清胰岛素样生长因子 - 1(IGF - 1)的浓度以及骨组织中转化生长因子 β1(TGF - β1)的表达 ,进一步探讨绝经后骨质疏松与细胞因子的相关性。方法 :将 6 0只10~ 12月龄的SD大鼠随机分成实验组和对照组 ,用摘除双侧卵巢的方法制备实验大鼠骨质疏松模型 ,实验组行摘除双侧卵巢术 ,对照组行假手术 ,仅切除卵巢周围的脂肪组织 ,应用HOLOGIC第 4代双能X线4 5 0 0W骨密度仪测量去卵巢不同时期 (术后 8,12 ,16周 )大鼠全身骨密度和腰椎感兴趣区域 (L1~L4)骨密度 ,并采用ELISA法和免疫组化的方法 ,检测去卵巢大鼠不同时期血清IGF - 1水平和骨组织中TGF - β1表达。结果 :全身骨密度 :去卵巢组大鼠在术后第 8周和第 12周 ,全身骨密度与对照组 (假手术组 )无明显差异(P >0 .0 5 ) ,在术后第 16周 ,开始出现全身骨密度降低 ,与对照组相比 ,有显著差异 (P <0 .0 1)。腰椎 (L1~L4)骨密度 :从术后第 8周~第 16周去卵巢腰椎骨密度均明显低于对照组 (P <0 .0 1)。血清IGF - 1浓度 :从术后第 8周~第 16周 ,去卵巢组血清IGF - 1浓度值均低于对照组 (P <0 .0 5 )。骨组织TGF - β1表达 :去卵巢组在术后第 8周骨组织中软骨细胞、成骨细胞、骨基质TGF - β1的表达程度与对照级有?
To observe the changes of bone mineral density (BMD), serum insulin - like growth factor - 1 (IGF - 1) and the expression of transforming growth factor β1 (TGF - β1) in osteoporosis rats at different stages, Loose and cytokine relatedness. Methods: Sixty SD rats aged 10 ~ 12 months were randomly divided into experimental group and control group. The experimental osteoporosis model was made by bilateral ovary removal. The experimental group received bilateral ovary surgery and the control group Undergoing sham operation, only the adipose tissue around the ovary was excised. Whole body BMD and lumbar spine were measured by HOLOGIC 4th generation dual-energy X-ray 4500W bone densitometry at different stages of ovariectomy (8, 12, 16 weeks after operation) (L1 ~ L4) bone mineral density. ELISA and immunohistochemistry were used to detect the serum IGF - 1 level and the expression of TGF - β1 in bone tissue of ovariectomized rats. Results: Whole-body BMD: There was no significant difference in total BMD between the ovariectomized group and the control group (P> 0.05) at the 8th week and the 12th week after operation in the ovariectomized group There was a significant difference (P <0.01) in total body mineral density reduction from the control group. BMD of lumbar vertebrae (L1 ~ L4): BMD of lumbar vertebrae from the 8th week to the 16th week after operation was significantly lower than that of the control group (P <0.01). Serum IGF - 1 levels: From the 8th week to the 16th week, the serum IGF - 1 levels in the ovariectomized group were lower than those in the control group (P <0.05). The expression of TGF - β1 in bone tissue: Compared with the control group, the expression level of TGF - β1 in chondrocytes, osteoblasts and bone matrix in the ovariectomized group at 8th week after operation.
