目的:探讨核因子-κB(NF-κB)“圈套”寡脱氧核苷酸(oligodeoxynucleitides,ODN)对子痫前期大鼠模型母胎界面炎症反应的影响。方法:①NF-κB“圈套”寡脱氧核苷酸(NF-κB decoy ODN)的设计、合成及转染;②研究NF-κB decoy ODN对子痫前期大鼠模型母胎界面的作用:Wistar孕鼠100只,随机分成正常对照组、子痫前期组、ODN组、脂质体组、错配ODN组,检测母胎界面的NF-κB表达和炎症反应。结果:子痫前期大鼠母胎界面NF-κB的表达明显增强,同时炎症反应较重。NF-κB decoy ODN能有效抑制母胎界面的炎症反应,而错配ODN却没有该效果。结论:NF-κB decoy ODN能有效抑制NF-κB介导的各类炎症因子的表达和合成,减轻子痫前期母胎界面的炎症反应。
Objective: To investigate the effect of nuclear factor-κB (NF-κB) and oligodeoxynucleitides (ODN) on the inflammatory reaction in the maternal-fetal interface of preeclampsia rats. Methods: ①The design, synthesis and transfection of NF-κB decoy ODN; ② The effect of NF-κB decoy ODN on maternal-fetal interface of preeclamptic rat model: Wistar 100 pregnant rats were randomly divided into normal control group, preeclampsia group, ODN group, liposome group and mismatch ODN group. The expression of NF-κB and inflammatory reaction in maternal-fetal interface were detected. Results: The expression of NF-κB in the maternal-fetal interface of preeclampsia was significantly increased and the inflammatory reaction was more serious. NF-κB decoy ODN can effectively inhibit the inflammatory reaction at maternal-fetal interface, whereas mismatched ODN did not. CONCLUSION: NF-κB decoy ODN can effectively inhibit the expression and synthesis of various inflammatory factors mediated by NF-κB, and reduce the inflammatory reaction at the interface of preeclampsia.
