目的探讨应用简化细胞分析软件分析角膜内皮细胞图像时,抽样细胞数量对分析结果可靠性的影响。设计临床试验研究。研究对象同一志愿者右眼的中央角膜内皮细胞图像30张。方法用非接触角膜内皮镜拍摄清晰的内皮细胞图像。应用细胞中心分析法对内皮细胞图像进行定量分析,每次分析时抽取不同数量的细胞,从10个细胞至90个细胞,每一图像分析9次。比较不同抽样细胞数量所得各组分析结果间的差异。主要指标角膜内皮细胞密度、细胞密度的变异系数。结果各组内皮细胞密度的变异系数随着点数细胞数量的增加,呈现逐渐下降并趋于稳定的趋势,点数细胞数10、20、30、40、50至90的细胞密度变异系数分别为9.5、6.3、4.6、4.4、4.3至3.8。点数10个和20个细胞的两组内皮细胞密度与其他各组比较均有显著性差异(P<0.05);点数30至90个细胞的各组间比较均无显著性差异。结论应用细胞中心分析法对角膜内皮细胞图像进行定量分析时,点数细胞数量应大于等于30个。(眼科,2007,16:421-424)
Objective To investigate the influence of the number of sampled cells on the reliability of analytical results when using simplified cell analysis software to analyze corneal endothelial cell images. Design clinical trial research. Thirty subjects underwent central corneal endothelial cell imaging on the right eye of the same volunteer. Methods Non-contact corneal endoscope for clear endothelial cell images. Endothelial cell images were quantitatively analyzed using a cell center assay with different numbers of cells from 10 cells to 90 cells for each analysis, 9 times for each image. Differences in the results of the analyzes of the groups obtained from the number of different sampled cells were compared. The main indicators of corneal endothelial cell density, coefficient of variation of cell density. Results The coefficient of variation of endothelial cell density in each group tended to decrease gradually and tended to be stable with the increase of the number of cells in each group. The coefficient of variation of cell density at the points of 10, 20, 30, 40, 50 to 90 was 9.5, 6.3, 4.6, 4.4, 4.3 to 3.8. There were significant differences in endothelial cell density between two groups of 10 and 20 cells (P <0.05). There was no significant difference between the groups of 30 to 90 cells. Conclusion Quantitative analysis of corneal endothelial cell images using cell-centric assay should have at least 30 cell numbers. (Ophthalmology, 2007, 16: 421-424)
