目的探讨防己黄芪汤治疗胰岛素抵抗的可能作用机制。方法将SD大鼠随机分为空白组、模型组、中药组及西药组各10只,除空白组外其余各组采用高脂饲料饲养12周建立胰岛素抵抗模型。造模成功后中药组按1 ml/100 g灌服防己黄芪汤(浓度0.6 g/ml),西药组按1 ml/100 g灌服吡格列酮悬浊液(浓度1 mg/ml),模型组及空白组灌服等容量蒸馏水,每日1次,各组连续给药2周。造模后检测各组大鼠葡萄糖输注率(GIR);给药后检测血清游离脂肪酸、脂联素、瘦素、抵抗素含量;油红O染色观察骨骼肌脂肪异位沉积程度;Western-blotting法测定骨骼肌腺苷酸活化蛋白激酶(AMPK)、p38丝裂原活化蛋白激酶(p38MAPK)蛋白含量。结果与空白组比较,模型组大鼠血清游离脂肪酸、瘦素、抵抗素含量及p38MAPK蛋白表达显著升高,脂联素含量、GIR、AMPK蛋白表达显著降低(P<0.05或P<0.01);与模型组比较,西药组、中药组游离脂肪酸、瘦素、抵抗素含量及p38MAPK蛋白表达显著降低,脂联素含量、AMPK蛋白表达显著升高(P<0.05或P<0.01)。油红O染色显示,与模型组比较,中药组和西药组大鼠骨骼肌红染颗粒减少,脂质沉积改善。结论防己黄芪汤可改善大鼠胰岛素抵抗脂质代谢紊乱,调节脂肪因子水平,其机制可能是通过调节AMPK信号转导通路蛋白水平实现的。
Objective To investigate the possible mechanism of anti-Jihuangqi Decoction in the treatment of insulin resistance. Methods SD rats were randomly divided into blank group, model group, TCM group and western medicine group, 10 rats in each group except the blank group. The rats in each group were fed with high-fat diet for 12 weeks to establish insulin resistance model. After the model was established, the Chinese medicine group was given 0.6g / ml Radix Astragali at a dosage of 1 ml / 100g, and the pioglitazone suspension (at a concentration of 1 mg / ml) The blank group was given the same volume of distilled water once a day for 2 weeks. After the model was established, the glucose infusion rate (GIR) of rats in each group was measured. The levels of free fatty acids, adiponectin, leptin and resistin in serum were detected after administration. The degree of ectopic deposition of skeletal muscle was observed by oil red O staining. The levels of AMPK and p38 MAPK in skeletal muscle were determined by Western blotting. Results Compared with the blank group, the levels of serum free fatty acid, leptin and resistin and the protein expression of p38MAPK in the model group were significantly increased (P <0.05 or P <0.01). The levels of adiponectin, GIR and AMPK were significantly decreased. Compared with model group, the contents of free fatty acids, leptin, resistin and p38MAPK protein in western medicine group and traditional Chinese medicine group were significantly decreased, and the content of adiponectin and AMPK protein were significantly increased (P <0.05 or P <0.01). Oil red O staining showed that compared with the model group, the Chinese medicine group and the western medicine group reduced the skeletal muscle red dye particles and improved the lipid deposition. Conclusion Fangji Huangqi Decoction can improve insulin resistance in rats with lipid metabolism disorders, regulation of adipokines levels, its mechanism may be through the regulation of AMPK signal transduction pathway protein levels achieved.
