目的探讨喉鳞状细胞癌(LSCC)组织中有丝分裂阻滞缺陷蛋白(MAD2)的表达及临床意义。方法采用免疫组织化学EliVision二步法检测55例LSCC组织、30例癌旁正常黏膜组织中MAD2蛋白的表达并分析其与LSCC临床病理特征的关系。结果 MAD2蛋白在LSCC组织中的阳性表达率为58.2%(32/55),低于癌旁正常黏膜组织86.7%(26/30),两者差异有统计学意义(P<0.01);中、低分化组MAD2阳性表达率为47.1%(16/34),低于高分化组76.2%(16/21),淋巴结转移组MAD2阳性表达率为35.0%(7/20),低于无淋巴结转移组71.4%(25/35),两组差异均有统计学意义(P均<0.05);MAD2阳性表达与患者性别、年龄、吸烟、肿瘤原发部位、T分期及临床分期无相关性(P均>0.05)。结论 MAD2蛋白在LSCC的发生、分化及转移中可起重要作用,可作为LSCC诊断及预后的参考指标之一。
Objective To investigate the expression and clinical significance of mitotic arrest defect protein (MAD2) in laryngeal squamous cell carcinoma (LSCC). Methods The immunohistochemical EliVision two-step method was used to detect the expression of MAD2 protein in 55 LSCC tissues and 30 adjacent normal mucosa tissues. The relationship between MAD2 protein expression and clinicopathological features of LSCC was analyzed. Results The positive expression rate of MAD2 protein in LSCC was 58.2% (32/55), which was lower than that in normal mucosa (86.7%, 26/30), the difference was statistically significant (P <0.01) The positive expression rate of MAD2 in poorly differentiated group was 47.1% (16/34), which was lower than 76.2% (16/21) in well differentiated group and 35.0% (7/20) in lymph node metastasis group, which was lower than that in non-lymph node metastasis (P <0.05). The positive expression of MAD2 had no correlation with the gender, age, smoking, primary tumor site, T stage and clinical stage (P All> 0.05). Conclusion MAD2 protein may play an important role in the occurrence, differentiation and metastasis of LSCC, which may be used as a reference index for the diagnosis and prognosis of LSCC.
