目的:用分光光度法研究门冬氨酸与2,3-二氯-5,6-二氰-1,4-苯醌(DDQ)的荷移反应。方法:门冬氨酸与DDQ于pH9.5的三酸缓冲溶液中,60℃水浴中加热15 min,可生成稳定的荷移络合物。结果:荷移络合物的组成比为1:1,λmax=342 nm,表观摩尔吸光系数ε=3.42×103L/mol.cm,在10~30μg/ml的范围内符合比尔定律,回收率为97.1%~101.7%,相对标准偏差在2.9%以内。结论:应用本方法测定门冬氨酸样品的含量,结果与文献方法一致。
OBJECTIVE: To study the charge transfer reaction of aspartate with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) by spectrophotometry. Methods: Aspartate and DDQ were hydrolyzed in pH9.5 aqueous solution at 60 ℃ for 15 min to produce a stable charge-transfer complex. Results: The molar ratio of charge-transfer complex was 1: 1, λmax = 342 nm, and the apparent molar absorptivity ε = 3.42 × 103 L / mol.cm. Beer’s law was obeyed in the range of 10 ~ 30 μg / 97.1% ~ 101.7%, the relative standard deviation was within 2.9%. Conclusion: This method is used to determine the content of aspartate samples. The results are consistent with the literature methods.
