目的:研究人肝脏DnaJ-like蛋白(human liver Dna J-like protein,HLJ1)在肝癌细胞中的表达,探讨HLJ1对肝癌细胞SK-Hep1生物学行为的影响。方法:首先,qRT-PCR和Western blot检测HLJ1在肝癌细胞中的表达情况。其次,过表达或干扰HLJ1后转染SK-Hep1细胞株,MTS和克隆形成实验检测细胞增殖能力的变化;流式细胞术检测细胞早期凋亡率;划痕实验检测细胞迁移能力;Transwell小室检测细胞侵袭能力的变化。结果:qRT-PCR和Western blot结果显示HLJ1的mRNA水平和蛋白水平在3种肝癌细胞系中均呈低表达。MTS和克隆形成结果显示HLJ1能抑制肝癌细胞的增殖。流式细胞术结果显示HLJ1能促进细胞早期凋亡。划痕和Transwell实验显示,HLJ1能抑制肝癌细胞的迁移和侵袭。结论:HLJ1在肝癌细胞系中低表达,且能抑制SK-Hep1细胞增殖、迁移和侵袭,促进细胞凋亡。
Objective: To study the expression of human liver Dna J-like protein (HLJ1) in hepatocellular carcinoma cells and to explore the effect of HLJ1 on the biological behavior of hepatocellular carcinoma cell line SK-Hep1. Methods: Firstly, the expression of HLJ1 in hepatocellular carcinoma cells was detected by qRT-PCR and Western blot. Secondly, SK-Hep1 cells were transfected overexpressing or interfering with HLJ1, MTS and colony formation assays were used to detect cell proliferation. Flow cytometry was used to detect cell early apoptosis rate. Scratch assay was used to detect cell migration ability. Transwell chamber assay Changes in cell invasiveness. Results: The results of qRT-PCR and Western blot showed that HLJ1 mRNA and protein levels were all low in all three cell lines. MTS and clone formation results show that HLJ1 can inhibit the proliferation of liver cancer cells. Flow cytometry results showed that HLJ1 can promote cell apoptosis in early stage. Scratches and Transwell experiments showed that HLJ1 can inhibit the migration and invasion of liver cancer cells. Conclusion: HLJ1 is low expressed in hepatocellular carcinoma cell lines, and can inhibit the proliferation, migration and invasion of SK-Hep1 cells and promote apoptosis.
