应用酶消化法体外培养猪气道上皮细胞 (AEC) ,加入一定浓度梯度的白细胞介素 - 1β (IL - 1β)和肿瘤坏死因子α (TNF-α) ,应用免疫细胞化学染色和原位杂交方法观测 AEC中 E钙粘着蛋白 (ECD)表达的变化。结果发现 :正常 AEC中 ECD表达于相邻上皮细胞间的胞膜上 ,其 m RNA也有一定含量。一定浓度梯度的 IL - 1β和 TNF-α作用 2 4h后 ,胞膜上 ECD表达减弱 ,而胞浆内 ECD表达在低浓度时升高 ,高浓度时降低 (P<0 .0 1) ,同时 ECD m RNA含量在低浓度时增多 ,而在高浓度时维持正常水平 (P>0 .0 5 )。结果提示 ,IL - 1β和 TNF-α在翻译或翻译后水平使膜上 ECD表达下调 ,参与了 AEC的损伤修复反应
Porcine airway epithelial cells (AECs) were cultured in vitro by enzymatic digestion. The results showed that: normal AEC ECD expression in the membrane between adjacent epithelial cells, m RNA also have a certain content. After a certain concentration gradient of IL - 1β and TNF - α for 24 hours, the expression of ECD on the membrane decreased, while the expression of ECD in cytoplasm increased at low concentration and decreased at high concentration (P <0.01) The content of ECD m RNA increased at low concentration and remained normal at high concentration (P> 0.05). The results suggest that IL - 1β and TNF - α at the translational or posttranslational level downregulate the expression of ECD on the membrane, which is involved in the damage repair reaction of AEC
