目的研究1,25-二羟维生素D3对角膜移植术后免疫排斥反应的影响。方法以45只SD大鼠为受体,15只Wistar大鼠为供体建立穿透性角膜移植模型。受体SD大鼠随机分成实验组、实验对照组和对照组,每组15只。实验组及实验对照组行同种异体角膜移植,对照组行自体角膜移植。术后0~13d实验组腹腔注射1,25-二羟维生素D31.0μg·kg-1·d-1,对照组和实验对照组注射灭菌花生油2ml/d;术后观察植片存活情况,并于术后14、21、30d每组随机处死5只大鼠行植片病理学检查和ELISA检测外周血IL-1β、IL-2、IL-8、IL-10。结果对照组角膜植片存活时间为(21.7±6.8)d,实验对照组为(11.2±2.5)d,实验组为(19.3±5.2)d,3组比较统计学差异非常显著(P<0.01);病理学检查示实验组淋巴细胞浸润和新生血管均较实验对照组少;实验组外周血中IL-1β、IL-2、IL-8含量与实验对照组相比下降,IL-10含量增高,统计学差异非常显著(P<0.01)。结论1,25-二羟维生素D3能在角膜移植后有效的保护植片,抑制排斥反应,延长植片存活时间。
Objective To investigate the effects of 1,25-dihydroxyvitamin D3 on immune rejection after corneal transplantation. Methods 45 penetrating keratoplasty models were established by using 45 SD rats as recipients and 15 Wistar rats as donors. Recipient SD rats were randomly divided into experimental group, experimental control group and control group, 15 rats in each group. Allogeneic corneal transplantation was performed in the experimental group and experimental control group, while autologous corneal transplantation was performed in the control group. The experimental group was injected intraperitoneally with 1,25-dihydroxyvitamin D31.0 μg · kg-1 · d-1 at 0 ~ 13 days after operation, while the control group and the experimental group were injected with sterilized peanut oil at 2 ml / d. Survival, Five rats were randomly sacrificed on the 14th, 21st and 30th day postoperatively for histopathology and ELISA for detection of IL-1β, IL-2, IL-8 and IL-10 in peripheral blood. Results The survival time of corneal graft was (21.7 ± 6.8) d in the control group, (11.2 ± 2.5) days in the experimental group and (19.3 ± 5.2) days in the experimental group. The statistical difference was significant (P <0.01) ; Pathological examination showed that the experimental group lymphocyte infiltration and neovascularization were less than the experimental control group; the experimental group of peripheral blood IL-1β, IL-2, IL-8 levels compared with the experimental control group, IL-10 content increased , The difference was statistically significant (P <0.01). Conclusion 1,25-dihydroxyvitamin D3 can effectively protect the graft after corneal transplantation, inhibit the rejection and prolong the survival time of the graft.
