目的:观察参麦注射液(SMI)对心衰犬心钠素(ANP)影响的动态变化。方法:将造模成功4周后的心衰犬20只随机分为对照组和SMI低、中、高剂量组,每组5只。对照组予生理盐水(0.9%NS)20 mL,低、中、高剂量组分别予SMI 0.517,1.034,1.551 mL.kg-1(用0.9%NS稀释至20 mL)iv qd×7。于给药前后检测心衰犬左室射血分数,并分别于给药前及给药后72h,144 h时点,取静脉血2 mL冰冻保存,用放射免疫分析法测定血浆ANP浓度。结果:SMI能够改善心衰犬的射血分数,中、高剂量作用明显;血浆心钠素水平下调,并呈现一定的量效、时效关系。结论:SMI对心衰犬射血分数的改善有剂量依赖趋势,对心钠素具有良性动态影响,这可能是其治疗心衰作用机制之一。
Objective: To observe the dynamic changes of the effect of Shenmai injection (SMI) on atrial natriuretic peptide (ANP) in heart failure dogs. Methods: Twenty heart failure dogs were randomly divided into control group and SMI low, middle and high dose groups, 5 in each group. The control group received 20 mL of normal saline (0.9% NS), and the low, medium, and high dose groups received SMI 0.517, 1.034, and 1.551 mL.kg-1 (diluted to 20 mL with 0.9% NS, respectively) iv qd×7. The left ventricular ejection fraction of heart failure dogs was measured before and after administration. Before and at the time points of 72 h and 144 h after drug administration, 2 mL of venous blood was collected and stored frozen. The plasma ANP concentration was determined by radioimmunoassay. RESULTS: SMI could improve the ejection fraction of heart failure dogs, and the effect of medium and high doses was obvious. The level of plasma atrial natriuretic peptide was down-regulated and showed a certain dose-effect and time-effect relationship. Conclusion: SMI has a dose-dependent trend in the improvement of ejection fraction of heart failure dogs and has a benign dynamic effect on atrial natriuretic peptide, which may be one of the mechanisms of SMI in the treatment of heart failure.
