目的研究Nod样受体热蛋白结构域相关蛋白3(NLRP3)在人脑胶质瘤组织中的表达,探讨NLRP3在人脑胶质瘤的发生、发展中的作用。方法按照2007年WHO中枢神经系统肿瘤分类及分级标准,将所收集的49例人脑胶质瘤组织根据其病理类型分为低级别组(20例)和高级别组(29例),并收集脑外伤后行颅内减压手术切除的正常脑组织10例做对照。应用免疫组织化学技术,并采用实时荧光定量逆转录聚合酶链反应(QRT-PCR)和免疫印迹技术,检测NLRP3在各组人脑胶质瘤及人正常脑组织中的表达。结果 NLRP3在正常脑组织中表达不明显,在低级别胶质瘤中表达较少,在高级别胶质瘤中表达明显。NLRP3 mRNA在正常脑组织(1.04±0.52)、低级别组(1.85±1.02)和高级别组(3.36±1.38)比较,差异有统计学意义(P<0.01)。人脑胶质瘤组织中NLRP3在mRNA水平及蛋白水平均高于正常脑组织(P<0.01)。NLRP3的表达与人脑胶质瘤的恶性程度呈正相关。结论人脑胶质瘤中存在NLRP3的异常表达,其表达量随胶质瘤恶性程度的升高而增加,NLRP3在人胶质瘤的发生和侵袭中可能发挥着重要的作用,检测NLRP3的表达有助于胶质瘤分化程度的判断。
Objective To investigate the expression of Nod-like receptor hot protein domain-related protein 3 (NLRP3) in human glioma and to explore the role of NLRP3 in the pathogenesis and progression of human glioma. Methods According to the WHO classification of the central nervous system tumors in 2007 and the classification criteria, 49 cases of human glioma were divided into low-grade group (20 cases) and high-grade group (29 cases) according to their pathological types and collected 10 cases of normal brain tissue resected by intracranial decompression after brain trauma were compared. The expression of NLRP3 in human gliomas and normal human brain tissues was detected by immunohistochemistry and real-time fluorescent quantitative reverse transcriptase polymerase chain reaction (QRT-PCR) and Western blotting. Results The expression of NLRP3 was not obvious in normal brain tissue, less in low grade glioma and significantly higher in high grade glioma. NLRP3 mRNA in normal brain tissue (1.04 ± 0.52), low grade group (1.85 ± 1.02) and high grade group (3.36 ± 1.38), the difference was statistically significant (P <0.01). NLRP3 mRNA and protein levels in human glioma tissue were higher than those in normal brain tissue (P <0.01). The expression of NLRP3 was positively correlated with the malignant degree of human glioma. Conclusion Abnormal expression of NLRP3 exists in human gliomas and its expression increases with the malignant degree of glioma. NLRP3 may play an important role in the pathogenesis and invasion of human glioma. The expression of NLRP3 may be detected Contribute to the judgment of glioma differentiation degree.
