用3×10~3pfu鼠巨细胞病毒(MCMV)对小鼠进行腹腔注射,发现感染后3d小鼠脾细胞总数下降,感染后5d脾中病毒效价达峰值时,脾细胞总数又回升。将感染后小鼠的T细胞进行体外培养,发现无论培养基中有无IL-2的存在,T细胞对ConA刺激的反应性都很低。采用流式细胞计检测细胞DNA含量,结果表明感染和未感染MCMV小鼠的新分离的T细胞中亚二倍体DNA含量分别为7.4%和4.4%,在ConA存在下培养12h后T细胞中亚二倍体DNA含量分别为26.2%和8.1%,培养24h后则分别为
The mice were injected intraperitoneally with 3 × 10 ~ 3 pfu murine cytomegalovirus (MCMV). The total number of splenocytes in mice was decreased 3 days after infection, and the total number of spleen cells rose again 5 days after infection. T cells from infected mice were cultured in vitro and found that T cells were less responsive to ConA stimulation regardless of the presence or absence of IL-2 in the medium. Flow cytometry was used to detect the DNA content of the cells. The results showed that the sub-diploid DNA content in newly isolated T cells from infected and non-infected MCMV mice was 7.4% and 4.4%, respectively.
