目的:研究补阳还五汤对异丙肾上腺素诱导大鼠心肌缺血的保护作用。方法:大鼠灌胃给予补阳还五汤,腹腔注射异丙肾上腺素制备心肌缺血模型,流式细胞技术检测外周血CD40的表达,免疫组织化学法观察心肌组织CD40L的表达,并测定血清LDH、CK、AST的活性。结果:心肌缺血模型组大鼠外周血CD40、心肌组织CD40L的表达及血清LDH、CK、AST的活性明显高于正常对照组(P<0.01),而补阳还五汤高、中剂量组大鼠外周血CD40、心肌组织CD40L的表达及血清LDH、CK、AST的活性较模型组显著降低(P<0.01)。结论:补阳还五汤对异丙肾上腺素诱导的大鼠心肌缺血具有保护作用,其作用可能与阻断CD40-CD40L炎症信号途径及降低血清LDH、CK与AST的活性有关。
Objective: To study the protective effect of Buyang Huanwu Decoction (GMP) on isoproterenol-induced myocardial ischemia in rats. Methods: The rat model of myocardial ischemia was established by intragastric administration of Buyang Huanwu Decoction and isoprenaline by intraperitoneal injection. The expression of CD40 in peripheral blood was detected by flow cytometry. The expression of CD40L in myocardium was observed by immunohistochemistry. LDH, CK, AST activity. Results: The expression of CD40L in myocardium and the level of serum LDH, CK and AST in myocardial ischemia model group were significantly higher than those in normal control group (P <0.01), while those in high and middle dose of Buyang Huanwu Decoction The expression of CD40L, LDH, CK and AST in peripheral blood of rats were significantly lower than those in model group (P <0.01). CONCLUSION: Buyang Huanwu Decoction can protect isoproterenol-induced myocardial ischemia in rats, which may be related to blocking the signal path of CD40-CD40L inflammation and decreasing the activities of serum LDH, CK and AST.
