赤芝是种重要的药用真菌,品种繁多,由于目前食用菌管理制度不完善,为稳定菌株质量迫切需要建立起快速鉴定赤芝菌株的有效办法。在对赤芝(Ganoderma lucidum)菌株进行SRAP多态性分析基础上,将属于某几个菌株的SRAP特异片段转化为稳定性较高的SCAR标记,共获得17个SCAR标记。聚类分析显示,供试的23个赤芝菌株在遗传距离0.63下分为4类,其中19个菌株在遗传距离0.50下聚成一类。将其中的9个SCAR标记线性组合,建立起赤芝菌株的多标记综合鉴别法,可对供试的23个菌株进行有效鉴别。由此可见,SCAR分子标记能很好地解释赤芝菌株间的亲缘关系,是种快速、稳定、准确鉴别赤芝菌株的方法。
Ganoderma lucidum is an important medicinal fungus, a wide variety, due to the current edible fungus management system is not perfect, there is an urgent need to establish an effective method for the rapid identification of Ganoderma strains to stabilize the quality of the strain. Based on SRAP polymorphism analysis of Ganoderma lucidum, 17 SRAR markers were obtained by transforming SRAP specific fragments belonging to several strains into SCAR markers with high stability. Cluster analysis showed that the 23 tested Ganoderma lucidum strains were divided into 4 groups at the genetic distance of 0.63, of which 19 strains clustered at a genetic distance of 0.50. Nine of the SCAR markers were linearly combined to establish a multi-marker comprehensive identification method for the Ganoderma lucidum strain, which can effectively identify the 23 strains tested. Thus, SCAR molecular markers can well explain the genetic relationship between Ganoderma strains, is a fast, stable and accurate method to identify Ganoderma strains.
