目的探讨亚砷酸钠(NaAsO_2)对PC12细胞的生长和3-巯基丙酮酸硫基转移酶(3MST)表达的影响。方法PC12细胞分别暴露于20、40、60、80、100μmol·L~(-1)NaAsO_212、24、36和48小时。倒置显微镜观察PC12细胞在药物处理后的形态变化。MTT法测定NaAsO_2在不同时间、不同浓度对PC12细胞存活率的影响;Western blot检测PC12细胞3MST的表达。结果亚砷酸钠引起细胞缩小、胞体变圆、细胞间隙变宽和死细胞增多等显著细胞形态学改变。40-100μmol·L~(-1)亚砷酸钠可以明显降低PC12细胞的存活率;3MST蛋白表达也显著减少,呈现剂量和时间依赖效应(P<0.05)。结论亚砷酸钠可抑制PC12细胞生长、诱导细胞损伤,其作用机制可能与下调3MST的表达有关。
Objective To investigate the effects of sodium arsenite (NaAsO_2) on the growth of PC12 cells and the expression of 3-mercapto-pyruvate sulfotransferase (3MST). Methods PC12 cells were exposed to 20, 40, 60, 80, 100μmol·L -1 NaAsO_212, 24, 36 and 48 hours, respectively. Inverted microscope was used to observe the morphological changes of PC12 cells after drug treatment. The effect of NaAsO_2 on the survival rate of PC12 cells was determined by MTT assay at different time and different concentrations. The expression of 3MST in PC12 cells was detected by Western blot. Results Sodium arsenite caused significant cell morphological changes such as cell shrinkage, round cell body, widening of cell gap and increase of dead cells. 40-100μmol·L -1 sodium arsenite can significantly reduce the survival rate of PC12 cells; the expression of 3MST protein also decreased significantly, showing dose-and time-dependent effects (P <0.05). Conclusion Sodium arsenite inhibits the growth of PC12 cells and induces cell injury. The mechanism may be related to the down-regulation of 3MST expression.
