本实验用相差显微镜、微分干涉差显微镜、扫描电镜和透射电镜,对肺泡巨噬细胞进行了观察;并用显微分光图像分析仪测定了细胞的酸性磷酸酶(AcP)、乳酸脱氢酶(LDH)和琥珀酸脱氢酶(SDH)含量。本文的结果表明,肺灌洗液中存在着两型不同的肺泡巨噬细胞。在相差显微镜下,呈现为明细胞和暗细胞;在微分干涉差显微镜和扫描电镜下,则呈现为球形细胞和扁平细胞。在扫描电镜下,球形细胞伸出较多的丝状突起,扁平细胞伸出较多的板状突起。在透射电镜下,球形细胞比扁平细胞含有更丰富的溶酶体。用定量细胞化学测定两型细胞的AcP、LDH、SDH含量,球形细胞明显高于扁平细胞。
In this experiment, alveolar macrophages were observed by phase-contrast microscope, differential interference microscope, scanning electron microscope and transmission electron microscope.
