目的 研究丹参对缺氧复氧后心室肌细胞L 型钙通道电流 (L Ica)的影响。方法 应用膜片钳全细胞记录技术。结果 经过 2 0min的缺氧以及 5min的复氧后 ,5 0、10 0、2 0 0mg/L丹参分别将L Ica的峰值从 (6 6 7 9± 15 8 7)pA减小至 (5 37 1± 12 1 3) pA、(4 97 2± 47 9) pA (P <0 0 5 ,n =5 )、(34 3 8± 73 1) pA(P <0 0 1,n =5 )。 结论 丹参能有效地减少缺氧复氧后心室肌细胞的L Ica ,这种作用可能是其抗心肌缺血以及缺血再灌性心律失常的机制之一。
Objective To study the effects of salvia miltiorrhiza on L-type calcium channel current (L Ica) in ventricular myocytes after hypoxia-reoxygenation. Methods Patch clamp whole cell recording technique was used. Results After 20 min of hypoxia and 5 min reoxygenation, 50, 100, 200 mg/L salvia miltiorrhiza reduce the peak value of L Ica from (6 679 9 18 8) pA to (5 37). 1 ± 12 1 3) pA, (4 97 2 ± 47 9) pA (P <0 0 5, n = 5), (34 3 8 ± 73 1) pA (P <0 0 1, n = 5). Conclusion Salvia miltiorrhiza can effectively reduce L Ica in ventricular myocytes after hypoxia-reoxygenation. This effect may be one of the mechanisms of its anti-myocardial ischemia and ischemia-reperfusion arrhythmia.
