目的探讨脂联素基因rs266729位点单核苷酸多态性与妊娠期糖尿病(diabetes gestational mellitus,GDM)的相关性。方法应用聚合酶链反应-限制性片段长度多态性(polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)技术,检测206例GDM患者(GDM组)其中GDM A2级患者25例(GDMA2组)和189例正常孕妇(对照组)脂联素基因rs266729位点多态性,并结合血糖、血脂等临床指标进行分析。结果 1对照组脂联素基因rs266729位点基因型CC、CG、GG频率分别为56.1%、38.6%、5.3%,GDM组分别为51.9%、40.8%、7.3%,两组比较差异无统计学意义(P>0.05);对照组等位基因C、G频率分别为75.4%、24.6%,GDM组分别为72.3%、27.7%,两组比较差异也无统计学意义(P>0.05)。2GDMA2组脂联素基因rs266729位点基因型CC、CG、GG频率分别为40.0%、52.0%、8.0%,与对照组比较差异无统计学意义(P>0.05);GDMA2组等位基因C、G频率分别为66.0%、34.0%,与对照组比较差异也无统计学意义(P>0.05)。3脂联素rs266729位点CC基因型组与CG+GG基因型组的年龄、OGTT三点血糖值、糖化血红蛋白、血脂、孕前及分娩前BMI等指标的比较,差异均无统计学意义。结论脂联素基因rs266729多态性可能与GDM发病无关,与GDM的严重程度亦无相关性。
Objective To investigate the association of adiponectin gene rs266729 SNP with gestational diabetes mellitus (GDM). Methods Twenty-five GDM patients (GDMA2 group) were detected in 206 GDM patients (GDM group) by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) And 189 cases of normal pregnant women (control group) adiponectin gene rs266729 polymorphism, combined with blood glucose, blood lipids and other clinical indicators were analyzed. Results The frequencies of CC, CG and GG of rs266729 locus in control group were 56.1%, 38.6% and 5.3% respectively, and those in GDM group were 51.9%, 40.8% and 7.3%, respectively. There was no significant difference between the two groups (P> 0.05). The frequencies of allele C and G in the control group were 75.4% and 24.6% respectively, and those in GDM group were 72.3% and 27.7% respectively. There was no significant difference between the two groups (P> 0.05). The frequencies of CC, CG and GG genotypes in 2GDMA2 group at rs266729 were 40.0%, 52.0% and 8.0%, respectively. There was no significant difference between the two groups (P> 0.05) G frequency were 66.0%, 34.0%, respectively, compared with the control group, the difference was not statistically significant (P> 0.05). There was no significant difference in age, OGTT three-point blood glucose, glycosylated hemoglobin, blood lipids, BMI before delivery and before delivery among CC genotype group and CG + GG genotype of adiponectin rs266729 locus. Conclusion Adiponectin rs266729 polymorphism may not be related to the pathogenesis of GDM, nor to the severity of GDM.
