目的观察转染了GSTθ后HCT116细胞系的生长、细胞周期的变化,以及HCT116细胞系生长对血清浓度的依赖性。方法通过脂质体Lipofectamine将质粒载体pcDNA3及pcDNA3-GSTθ分别转染HCT116细胞获得稳定转染株,通过RT-PCR检测GSTθ的表达,细胞计数绘制细胞生长曲线,流式细胞仪检测细胞周期变化,MTT法检测细胞生长对血清浓度的依赖。结果GSTθ转染后抑制了HCT116细胞的生长,并且是通过G2/M和S期阻滞而导致的,GSTθ还增加了HCT116细胞对血清的依赖性。结论GSTθ可降低HCT116细胞的恶性程度,使其对血清及营养物质的需求增加。
Objective To observe the growth of HCT116 cell line after transfection with GSTθ, the change of cell cycle and the dependence of HCT116 cell line on serum concentration. Methods Plasmid vector pcDNA3 and pcDNA3-GSTθ were transfected into HCT116 cells by Lipofectamine respectively. The expression of GSTθ was detected by RT-PCR, the cell growth curve was drawn by cell counting, the cell cycle was detected by flow cytometry, MTT assay was used to determine the dependence of cell growth on serum concentration. Results GST theta inhibited the growth of HCT116 cells and was arrested by G2 / M and S arrests. GSTθ also increased the serum dependence of HCT116 cells. Conclusions GSTθ can reduce the malignant degree of HCT116 cells and increase its demand for serum and nutrients.
