目的探讨GOLPH3在食管癌转移中的作用。方法通过划痕实验、Transwell侵袭实验和3D培养等方法,研究GOLPH3对KYSE-140细胞迁移和侵袭能力的影响。用q RT-PCR法检测CYR61,CD44与Snail这三个上皮-间质转化(epithelial-mesenchymal transition,EMT)相关分子与GOLPH3的关系,初步探索GOLPH3对食管鳞癌细胞转移能力的影响。结果 GOLPH3对食管鳞癌细胞的迁移和侵袭有促进作用,且CYR61,CD44与Snail在食管鳞癌细胞中的表达水平与GOLPH3的表达呈正相关。结论 GOLPH3高表达可促进食管鳞癌细胞的上皮-间质转化,从而促进食管癌的转移和进展,发挥癌基因作用。
Objective To investigate the role of GOLPH3 in the metastasis of esophageal cancer. Methods The effects of GOLPH3 on the migration and invasion of KYSE-140 cells were studied by scratch test, Transwell invasion assay and 3D culture. QRT-PCR was used to detect the relationship between GOLPH3 and CYR61, CD44 and Snail, and to explore the effect of GOLPH3 on the metastasis of esophageal squamous cell carcinoma. Results GOLPH3 promoted esophageal squamous cell carcinoma cell migration and invasion, and the expression of CYR61, CD44 and Snail in esophageal squamous cell carcinoma was positively correlated with the expression of GOLPH3. Conclusion GOLPH3 overexpression promotes the epithelial-mesenchymal transition of esophageal squamous cell carcinoma, which promotes the metastasis and progression of esophageal carcinoma and plays an oncogene role.
