目的 测定家兔实验性急性高眼压模型视网膜谷氨酸 (glutamate,GL U)、天门冬氨酸 (as-partate,ASP)、γ-氨基丁酸 (γ- aminobutyric acid,GABA)和甘氨酸 (glycine,GL Y)含量的动态变化。 方法 4 0只家兔分为 8组 ,每组 5只兔 ,每兔随机选一只眼作为实验眼。 1~ 6组为实验组 ,实验眼在 12 0mm Hg(1mm Hg=0 .133k Pa)高压前房灌注持续 15、30、4 5 min和高压灌注持续 4 5 min后恢复正常眼压15、30、4 5 min时摘除眼球。实验对照组仅作前房穿刺后即摘除眼球。空白对照组直接处死动物摘除眼球。利用邻苯二甲醛柱前衍生法 ,高效液相色谱仪测定各组兔眼视网膜氨基酸的含量。 结果 高眼压灌注持续 15 min组 ,视网膜 GL U水平较实验对照组显著升高 (t=5 .9814 ,P=0 .0 0 0 3) ;恢复正常眼压 15 min组 ,GL U含量再次显著升高 ,与实验对照眼比较差异显著 (t=2 .5 970 ,P=0 .0 318)。 ASP、GABA和 GL Y在实验过程中未发现有统计意义的变化。前房穿刺实验对照组与空白对照组的 GL U含量比较无差别。 结论 急性高眼压早期以及恢复正常眼压早期均可导致视网膜 GL U含量显著升高。
Objective To determine the effects of glutamate (GL U), as-partate (ASP), γ-aminobutyric acid (GABA) and glycine glycine, GL Y) content of the dynamic changes. Methods 40 rabbits were divided into 8 groups with 5 rabbits in each group. One rabbit was randomly selected as the experimental eye. Groups 1 to 6 were experimental groups. The experimental eyes underwent 15,30,4 5 minutes of high pressure anterior chamber perfusion at 120mm Hg (1mm Hg = 0.133k Pa) and 15, 30 , 4 5 min removal of the eye. Experimental control group only for anterior chamber puncture after removal of the eye. The blank control group was sacrificed directly to remove the eyeballs. The contents of amino acids in the retina of rabbits were detected by HPLC with POM column and high performance liquid chromatography. Results After intraocular pressure (IOP) infusion for 15 min, GL U level in retina was significantly higher than that in experimental group (t = 5 .9814, P = 0.00003) (T = 2.4970, P = 0.0183), which was significantly higher than that of experimental control eyes. ASP, GABA and GLY in the experiment found no statistically significant changes. The anterior chamber puncture experimental control group and the blank control group GL U content no difference. Conclusion Both acute early intraocular pressure (IOP) and early recovery of normal intraocular pressure (IOP) can lead to a significant increase of GL U in the retina.
