目的探讨补阳还五汤对同型半胱氨酸(homocysteine,Hcy)致动脉粥样硬化(AS)的保护作用与核因子(NF)-κB的关系。方法给予ApoE-/-小鼠Hcy(0.1 g.kg-1)灌胃8周后造成动脉粥样硬化(AS)模型,治疗组给予补阳还五汤(5,10,20g.kg-1)进行灌胃。8周后HE染色观察和测量主动脉根部AS斑块面积、血管壁面积,检测主动脉NF-κB活性、活性氧(ROS)含量、I-κBα蛋白表达变化。结果模型组AS病变明显,与正常对照组相比,主动脉组织中ROS含量显著升高(P<0.01),I-κBα蛋白表达明显降低,NF-κB活性显著激活;不同剂量的补阳还五汤改善AS病变的同时显著降低血管组织中ROS含量、促进I-κBα蛋白表达升高,抑制NF-κB的活化。结论补阳还五汤可能通过抑制NF-κB的活化达到抗Hcy所致动脉硬化的作用。
Objective To investigate the protective effect of Buyang Huanwu decoction on homocysteine (Hcy) -induced atherosclerosis (AS) and its relationship with nuclear factor (NF) -κB. Methods Atherosclerosis (AS) model was induced by intragastric administration of Hcy (0.1 g · kg-1) in ApoE - / - mice for 8 weeks. The treatment group was given BYHWD (5, 10, ) For gavage. After 8 weeks, the area of aortic root plaque, the area of vascular wall, the activity of NF-κB, the content of reactive oxygen species (ROS) and the expression of I-κBα in aortic root were observed and measured by HE staining. Results Compared with the normal control group, the content of ROS in the aortic tissue increased significantly (P <0.01), the expression of I-κBα protein decreased and the activity of NF-κB markedly increased in model group. Compared with normal control group, Wu Tang can improve the pathological changes of AS and significantly reduce the content of ROS in vascular tissue, promote the expression of I-κBα protein and inhibit the activation of NF-κB. Conclusion Buyang Huanwu decoction may inhibit the activation of NF-κB to anti-Hcy-induced atherosclerosis.
