目的观察缺血性脑损伤后大鼠缺血皮质区microRNA 210的表达变化规律,探讨mir-210对脑缺血后血管新生的可能影响。方法实验分为缺血后1d、3d、7d组和假手术组(n=5),缺血组采用线栓法制作大脑中动脉阻塞模型,于缺血后1、3、7d处死大鼠,取缺血皮质区用于实验;假手术组仅暴露动脉。提取各实验组脑缺血皮质区的miRNA,Real time PCR比较各实验组mir-210的表达水平。结果脑缺血后mir-210表达显著上调,缺血后1d、3d、7d分别为假手术组的7.2±0.56、20.1±0.87和20.3±0.76倍(P<0.05)。结论脑缺血后mir-210表达显著上调,mir-210可能在促进脑缺血后血管新生过程中起重要作用。
Objective To observe the changes of microRNA 210 expression in ischemic cortex after ischemic brain injury in rats and explore the possible influence of mir-210 on angiogenesis after cerebral ischemia. Methods The experiment was divided into 1d, 3d, 7d group and sham operation group (n = 5) after ischemia. The middle cerebral artery occlusion model was established by thread occlusion in the ischemic group. Rats were sacrificed at 1, Ischemic cortex regions were used for the experiment; the sham-operated group only exposed arteries. The miRNAs of cerebral ischemia cortex in each experimental group were extracted, and the expression of mir-210 in each experimental group was compared by Real time PCR. Results The expression of mir-210 was significantly increased after cerebral ischemia. The levels of mir-210 in the ischemic group were 7.2 ± 0.56, 20.1 ± 0.87 and 20.3 ± 0.76 folds (P <0.05) on the 1st, 3rd and 7th day after ischemia, respectively. Conclusion The expression of mir-210 after cerebral ischemia is significantly increased, and mir-210 may play an important role in promoting angiogenesis after cerebral ischemia.
