目的 探讨 Fasl(Fas- L igand)在 ABM(activated bone marrow)杀伤肿瘤细胞效应中的作用。方法 应用抗 CD3 单克隆抗体体外激活骨髓细胞 ,采用免疫荧光标记、流式细胞仪检测 ,观察了不同条件下 ABM细胞表面 Fas L 的表达。结果 未激活的骨髓细胞不表达 Fas L,r IL- 2激活后可见 Fas L 的表达 (7.0 4 %± 3 .4 4% ) ,抗 CD3单抗可促进其表达 (11.84 %± 4 .80 % ) ,三组比较具有显著性差异 (P<0 .0 0 5 )。结论 ABM杀伤肿瘤细胞的机制与 Fas/ Fas L 介导的肿瘤细胞凋亡有关。
Objective To investigate the role of Fasl (Fas-Ligand) in the killing of tumor cells by activated bone marrow (ABM). Methods The anti-CD3 monoclonal antibody was used to activate bone marrow cells in vitro. Immunofluorescence staining and flow cytometry were used to observe the expression of Fas L on the surface of ABM cells under different conditions. Results The unactivated bone marrow cells did not express Fas L, and the expression of Fas L was observed after activation of r IL-2 (7.04% ± 3.44%). The anti-CD3 monoclonal antibody promoted its expression (11.84 % ± 4.80 % ), There was a significant difference between the three groups (P<0.05). Conclusion The mechanism of ABM killing tumor cells is related to Fas/Fas L-mediated tumor cell apoptosis.
