目的研究黄芩苷磷脂复合物溶液单侧鼻腔给药后黄芩苷在大鼠体内血浆与左、右嗅球,左、右脑及小脑中的药物分布,并评价其脑靶向性。方法大鼠静注黄芩苷磷脂复合物溶液,单侧(左侧)鼻腔给予黄芩苷磷脂复合物溶液后,采用HPLC分别测定血浆中及各脑组织中黄芩苷的浓度。结果药物经左侧鼻腔给药后,最高药物浓度出现在左嗅球,其次为右嗅球,接下来分别为左脑、右脑及小脑。嗅区转移率分别为100%、100%、75.06%、62.39%、65.11%。结论黄芩磷脂复合物经鼻腔给药后,黄芩苷可由“鼻-脑”通路实现药物的脑靶向分布。
OBJECTIVE To study the drug distribution of baicalin in plasma and left and right olfactory bulb, left and right brain and cerebellum of rats after intranasal administration of baicalin phospholipid complex solution, and evaluate its brain targeting. Methods After the baicalin phospholipid complex solution was intravenously administered to rats, unilateral (left) intranasal administration of baicalin phospholipid complex solution was used to determine the concentration of baicalin in plasma and brain tissue respectively. Results After the drug was administered to the left nasal cavity, the highest drug concentration appeared in the left olfactory bulb, followed by the right olfactory bulb, followed by the left brain, right brain and cerebellum, respectively. Olfactory area transfer rates were 100%, 100%, 75.06%, 62.39%, 65.11%. Conclusions Baicalin can achieve the brain-targeted distribution of drugs by the “nasal-brain” pathway after intranasal administration of the baicalin phospholipid complex.
