目的微小RNA(microRNA,miRNA)表达或功能异常在肿瘤发生发展中发挥重要作用,微小RNA-762(microRNA-762,miR-762)在胃癌发生发展中的作用目前仍罕见报道。故本研究探讨miR-762在胃癌组织中的表达、临床意义及生物学功能。方法采用qRT-PCR检测新疆医科大学附属自治区中医院普外科2009-01-01-2014-12-31 60例胃癌及相应癌旁组织中miR-762的表达,并分析其与胃癌患者临床病理特征的相关性,并利用Transwell检测miR-762对胃癌细胞迁移和侵袭能力的影响。结果 miR-762在胃癌组织中的表达水平为1.98±0.23,显著高于相应癌旁组织的1.00±0.04,P=0.003;miR-762在转移组胃癌组织中的表达水平为2.89±0.23,显著高于未转移组的1.76±0.05,t=25.45,P<0.05。miR-762高表达与淋巴结转移(P<0.001)及TNM分期(P=0.006 1)有显著相关性。过表达miR-762能够显著增强胃癌细胞的迁移和侵袭能力,敲低miR-762显著降低胃癌细胞迁移和侵袭能力。结论 miR-762表达增高与胃癌患者的不良临床病理特征相关。miR-762能够增强胃癌细胞的迁移和侵袭能力。故miR-762在胃癌的发生发展中发挥重要作用。
The expression or dysfunction of microRNAs (miRNAs) plays an important role in tumorigenesis. The role of microRNA-762 (miR-762) in the development of gastric cancer is still rarely reported. Therefore, this study to explore the expression of miR-762 in gastric cancer, clinical significance and biological function. Methods qRT-PCR was used to detect the expression of miR-762 in 60 cases of gastric cancer and corresponding paracancerous tissues, and to analyze its clinical and pathological features Transwell was used to detect the effect of miR-762 on the migration and invasion of gastric cancer cells. Results The expression level of miR-762 in gastric cancer was 1.98 ± 0.23, significantly higher than 1.00 ± 0.04 in the corresponding paracancerous tissues (P = 0.003). The expression level of miR-762 in metastatic gastric cancer was 2.89 ± 0.23 Higher than the non-metastasis group 1.76 ± 0.05, t = 25.45, P <0.05. miR-762 overexpression was significantly associated with lymph node metastasis (P <0.001) and TNM stage (P = 0.006 1). Overexpression of miR-762 significantly enhanced gastric cancer cell migration and invasion, and knockdown of miR-762 significantly reduced gastric cancer cell migration and invasion. Conclusion The increased expression of miR-762 correlates with poor clinical and pathological features in patients with gastric cancer. miR-762 enhances the ability of gastric cancer cells to migrate and invade. Therefore, miR-762 plays an important role in the occurrence and development of gastric cancer.
