目的研究大黄酸(RH)和大黄素(EM)与过氧化氢酶(CAT)的相互作用。方法采用荧光光谱法、紫外光谱法和圆二色谱法进行研究。结果确定了静态猝灭和非辐射能量转移是导致CAT荧光猝灭的主要原因,圆二色谱表明CAT中α-螺旋含量改变,热力学参数表明了药物分子与CAT之间的作用力类型。结论药物分子与CAT之间有较强的结合作用,以疏水作用为主,反应自发进行,其中EM对CAT作用较强。
Objective To study the interaction between rhein (RH) and emodin (EM) and catalase (CAT). Methods Fluorescence spectroscopy, UV spectroscopy and circular dichroism were studied. The results showed that static quenching and non-radiative energy transfer were the main causes of fluorescence quenching of CAT. Circular dichroism showed that the content of α-helix changed in CAT, and the thermodynamic parameters indicated the type of force between CAT and CAT. Conclusions There is a strong binding between drug molecules and CAT, mainly by hydrophobic interaction, and the reaction proceeds spontaneously. The effect of EM on CAT is stronger.
