为探讨不同X线处理对脑胶质瘤细胞增殖的影响,选用人脑星形细胞瘤SWO-38系,采用MTT法观察X线对胶质瘤细胞生长的影响,ABC法检测增殖细胞核抗原(PCNA)基因表达的变化。结果显示:①不同剂量(5,10,15,20,25Gy)X线对胶质瘤细胞生长具有明显的抑制作用,生长曲线低平,照射后48h的抑制率分别为33%、47%、51%、58%和66%;②不同剂量率(80,160,240,320,400cGy/min)X线照射后48h的抑制率分别为58%、54%、47%、25%和30%;③超分割照射后48h的抑制率为62%,而单次照射组为50%;④不同剂量(5,10,15,20,25Gy)X线照射后PCNA-标记率(LI)分别为91%,87%,81%,78%,69%,PCNA-LI与存活细胞OD值呈显著性正相关关系(P<0.05)。说明①SWO细胞生长曲线、抑制率与X线剂量、剂量率和照射方式之间密切相关;②PCNA-LI与存活细胞OD值呈正相关关系,PCNA是胶质瘤细胞增殖的标志之一。
In order to investigate the effect of different X-ray treatment on the proliferation of glioma cells, the human astrocytoma SWO-38 line was selected. The effect of X-ray on the growth of glioma cells was observed by MTT method. The proliferative cell nuclear antigen PCNA) gene expression changes. The results showed that: (1) X-ray at different doses (5, 10, 15, 20, 25 Gy) significantly inhibited the growth of glioma cells and the growth curve was leveled with the inhibition rates of 33%, 47% 51%, 58%, and 66% respectively; ②The inhibitory rates at different dose rates (80,160,240,320,400cGy / min) 48h after radiotherapy were 58%, 54%, 47%, 25% and 30% ; ③ The inhibition rate of 48h after superfractionated irradiation was 62%, while that of single irradiation group was 50%. ④ The PCNA-labeling rate (LI) after X-ray irradiation at different doses (5,10,15,20,25Gy) 91%, 87%, 81%, 78%, 69%. There was a significant positive correlation between PCNA-LI and surviving cell OD (P <0.05). There was a positive correlation between PCNA-LI and OD value of viable cells, and PCNA was one of the markers of glioma cell proliferation.
