目的 :观察血管紧张素受体拮抗剂氯沙坦对糖尿病大鼠肾脏功能和肾组织结构的影响。方法 :雄性Wis tar大鼠分为正常对照组 (C)、糖尿病对照组 (D)、糖尿病氯沙坦治疗组 (DL)。于实验第 6、12和 2 4周 ,测定各组大鼠的血糖、糖化血红蛋白、肌酐清除率及尿蛋白排泄率 ,并对肾脏标本进行了光镜、电镜观察 ,同时测量各组大鼠的平均肾小球体积。结果 :于第 6周、12周、2 4周 ,糖尿病对照组大鼠的上述指标与C组相比 ,均明显升高 (P <0 .0 5 ) ,而DL组尿蛋白排泄率均低于同时期的D组 (P <0 .0 5 )。DL组肾小球体积显著低于D组(P <0 .0 5 )。电镜检查发现D组基底膜显著增厚 ,系膜和系膜细胞显著增生 ,DL组上述改变明显减轻。结论 :氯沙坦能够延缓糖尿病大鼠肾脏病变的进程 ,保护肾功能。
Objective: To observe the effects of losartan, an angiotensin receptor antagonist, on renal function and renal structure in diabetic rats. Methods: Male Wis tar rats were divided into normal control group (C), diabetic control group (D) and diabetic losartan treatment group (DL). The blood glucose, glycosylated hemoglobin, creatinine clearance rate and urinary protein excretion rate of rats in each group were measured at the 6th, 12th and 24th weeks of the experiment, and the kidney specimens were observed under light and electron microscopes, and the rats’ Average glomerular volume. Results: In the 6th week, 12th week and 24th week, the indexes in the diabetic control group were significantly higher than those in the C group (P <0.05), while the urinary protein excretion rate in the DL group was significantly lower In the same period of Group D (P <0. 05). Glomerular volume in DL group was significantly lower than that in D group (P <0.05). Electron microscopy showed that the basement membrane in group D was significantly thicker, and mesangial and mesangial cells were significantly proliferated. The above changes in DL group were significantly reduced. Conclusion: Losartan can delay the progression of renal disease in diabetic rats and protect renal function.
