目的 以二烯丙基二硫 (DADS)诱导人急性早幼粒细胞性白血病HL60细胞分化为模型 ,利用双向凝胶电泳 ( 2 -DE)技术分析比较DADS诱导HL60细胞分化的蛋白表达差异。方法 细胞形态学观察与NBT还原反应结合验证HL60细胞的分化 ;用双向电泳技术分析蛋白表达差异。结果 分化后细胞形态发生明显变化 ,细胞核变小 ;NBT还原能力明显增强。双向电泳技术分析筛选了 32个差异点。结论 双向电泳技术可直接反映细胞蛋白表达的变化 ,DADS诱导HL60细胞分化后导致部分蛋白的表达增加和降低
OBJECTIVE: To differentiate human acute promyelocytic leukemia HL60 cells induced by diallyl disulfide (DADS), and analyze the difference of DADS-induced HL60 cell differentiation by two-dimensional gel electrophoresis (2-DE). Methods Cell morphology and NBT reduction were used to verify the differentiation of HL60 cells. The protein expression was analyzed by two-dimensional electrophoresis. Results After differentiation, the morphology of cells changed obviously and the nucleus became smaller. The NBT reduction ability was obviously enhanced. Two-dimensional electrophoresis analysis identified 32 differences. Conclusion Two-dimensional electrophoresis can directly reflect the changes of cell protein expression, DADS-induced differentiation of HL60 cells lead to increased and decreased part of the protein expression
