目的探讨葡萄籽提取物原花青素(PA)对人肝癌细胞SMMC-7721细胞的诱导分化作用。方法用不同剂量的PA与SMMC27721细胞共同培养,以MTT法检测细胞增殖和PA对细胞增殖的抑制作用;镜下观察细胞形态改变;采用RT-PCR方法检测细胞甲胎蛋白mRNA表达,金氏法检测细胞碱性磷酸酶活性的变化。结果 PA在20~60μg/ml的浓度范围均能抑制细胞增殖,且抑制率随浓度的增加而增高(P<0.05),细胞形态和结构向正常肝细胞方向逆转,40μg/ml和60μg/ml的PA均可使细胞甲胎蛋白mRNA表达量明显下降,碱性磷酸酶活性明显升高。结论 PA对人肝癌SMMC27721细胞具有抗增殖和促分化作用。
Objective To investigate the effect of grape seed extract procyanidin (PA) on the differentiation of human hepatoma SMMC-7721 cells. Methods PA and SMMC27721 cells were co-cultured with different doses of PA and MTT assay was used to detect the cell proliferation and the inhibitory effect of PA on cell proliferation. The morphology of cells was observed under microscope. The mRNA expression of alpha-fetoprotein was detected by RT- Changes in cell alkaline phosphatase activity were measured. Results PA inhibited cell proliferation in the concentration range of 20 ~ 60μg / ml, and the inhibitory rate increased with the increase of concentration (P <0.05). The morphology and structure of PA reversed to the normal hepatocytes, 40μg / ml and 60μg / ml Of PA can make cell alpha-fetoprotein mRNA expression decreased significantly, alkaline phosphatase activity was significantly increased. Conclusion PA has antiproliferative and differentiation effects on human hepatoma SMMC27721 cells.
