对2005~2008年深圳地区肠道病毒71型进行基因特征研究。用肠道病毒71型特异性引物进行RT-PCR,并扩增EV71的VP1基因,所得的核苷酸序列与肠道病毒71型A、B、C基因型代表株的核苷酸序列比较,并且用DNASTAR、BioEdit和Mega 3.1软件进行系统进化分析。结果表明17株病毒与C4a亚群代表株接近,VP1区核苷酸同源性在95.3%~99.4%之间,1株病毒与C4b亚群代表株较接近,核苷酸同源性在93.0%~95.6%之间;18株病毒与A、B基因型代表株比较差异较大,VP1区核苷酸同源性为81.8%~86.0%,18株病毒之间的VP1核苷酸同源性为92.5%~100%。深圳地区EV71的流行株均属于C4基因型,但以C4a亚群为主,还存在C4b亚群。
Gene characteristics of enterovirus 71 in Shenzhen area from 2005 to 2008 were studied. RT-PCR was performed with Enterovirus 71-specific primers and the VP1 gene of EV71 was amplified. The nucleotide sequence of the VP1 gene was compared with the nucleotide sequence of the representative strain of enterovirus 71 type A, B, C genotypes. Phylogenetic analysis was performed using DNASTAR, BioEdit and Mega 3.1 software. The results showed that 17 strains of viruses were close to the representative C4a subunit strain, the nucleotide homology of VP1 was between 95.3% and 99.4%, and one strain of virus was closer to that of C4b subunit. The nucleotide homology was 93.0 % ~ 95.6%. The 18 strains of virus showed significant differences from the representative strains of A and B genotypes, the nucleotide homology of VP1 was 81.8% -86.0%, and the VP1 nucleotide homology Sex is 92.5% ~ 100%. Epidemic strains of EV71 in Shenzhen belonged to genotype C4, but C4a subunit was dominant, while C4b subunit was still present.
