目的:建立一种稳定的、可操作性强的病证结合大鼠帕金森病(PD)模型,以满足研究药物对PD治疗评价的需求。方法:采用6-羟基多巴胺脑部立体注射法构建PD动物模型,同时设立正常对照组、假手术组、模型组、药物(复方地黄方)组,从病因、症状、客观指标、药物反证等方面对PD大鼠模型进行综合判断,验证该模型的中医证候属性。结果:模型组大鼠的神经行为学、氧化应激、TH m RNA及蛋白、阳性细胞等指标与正常对照组比较,差异均有统计学意义(P<0.01)。药物组可明显改善大鼠的主观症状和上述客观指标,差异有统计学意义(P<0.01)。结论:6-羟基多巴胺大鼠PD模型的中医证候属性是风阳内动证。
OBJECTIVE: To establish a stable and maladaptive disease syndrome combined with Parkinson’s disease (PD) model in rats to meet the needs of research evaluation of PD. Methods: PD animal models were established by 6-OHDA injection and normal control group, sham operation group, model group and drug (Compound Dihuang Recipe) were established at the same time. The etiology, symptom, objective index and anti-evidence Comprehensive judgment of PD rat model to verify the TCM syndrome attributes of the model. Results: The neurobehavioral, oxidative stress, TH m RNA, protein, positive cells and other indexes in model group were significantly different from those in normal control group (P <0.01). Drug group can significantly improve the subjective symptoms of rats and the above objective indicators, the difference was statistically significant (P <0.01). Conclusion: TCM syndromes of 6-hydroxydopamine in PD model is a positive-yang-yang motor.
