WRI1基因在控制从碳水化合物到脂类的转化过程中起着重要的作用。通过qRT-PCR方法分析了JcWRI1在麻疯树不同组织中的表达特性,结果表明:JcWRI1主要在种子中表达。同时克隆了麻疯树JcWRI1基因起始密码子上游2 000 bp序列。序列分析表明,该序列不仅包含有TATAbox和CAAT-box基本元件,而且含有胚乳表达所需的顺式调控元件、脱落酸应答相关顺式作用元件、光响应元件及各种胁迫响应元件。将该启动子与GUS报告基因连接构建表达载体,并转化水稻。GUS组织化学染色分析结果表明,JcWRI1启动子驱动GUS基因在根、茎、叶中有很低的表达,然而在种子中具有较高的表达活性。表明该启动子主要在种子中表达,并受多种因子的调控。
The WRI1 gene plays an important role in controlling the conversion from carbohydrates to lipids. The expression characteristics of JcWRI1 in different tissues of Jatropha curcas were analyzed by qRT-PCR. The results showed that JcWRI1 was mainly expressed in seeds. At the same time, a 2 000 bp sequence upstream of the start codon of JcWRI1 gene in Jatropha curcas was cloned. Sequence analysis showed that the sequence contains not only the basic elements of TATAbox and CAAT-box, but also the cis-regulatory elements required for endosperm expression, the cis-acting elements related to abscisic acid responses, light-responsive elements and various stress response elements. The promoter was ligated with the GUS reporter gene to construct an expression vector and transformed into rice. The results of GUS histochemical staining showed that GUS gene driven by JcWRI1 promoter had low expression in roots, stems and leaves, but had higher expression activity in seeds. This promoter is mainly expressed in seeds and is regulated by many factors.
