目的根据线粒体Cytb基因部分序列,对从入境集装箱中截获的疑似鼠样本进行鉴定种类。方法提取样本中的DNA,采用PCR方法进行线粒体Cytb基因扩增,对目的片段测序,通过blast比对进行同源性比对。从Genbank获取宁波口岸常见8个鼠种24个样本的Cytb基因序列作为参照,利用生物信息学软件Mega5.0进行系统进化分析。结果样本DXR1110与小家鼠同源性最高,DXR1203与褐家鼠同源性最高,系统进化树显示不同鼠种间形成独立分支。结论Cytb基因分析是进行鼠种鉴定的有效手段。
Objective To identify the suspected murine samples intercepted from inbound containers based on the mitochondrial Cytb gene partial sequence. Methods The DNA in the samples was extracted and the mitochondrial Cytb gene was amplified by PCR. The target fragments were sequenced and their homologies were compared by blast comparison. The Cytb gene sequences of 24 samples from 8 portraits of Ningbo Port at Ningbo Port were obtained from Genbank. The phylogenetic analysis was made using the bioinformatics software Mega5.0. Results The sample DXR1110 had the highest homology with the Mus musculus, DXR1203 had the highest homology with the brown rat and the phylogenetic tree showed that there was an independent branch between different species. Conclusion Cytb gene analysis is an effective method for identification of murine species.
