目的研究霉茶总黄酮对原发性高血压大鼠(SHRs)胸主动脉功能的影响及其作用机制。方法 SHRs随机分为模型组,总黄酮低(90 mg/kg)、中(180 mg/kg)、高剂量(360 mg/kg)组和复方罗布麻片(50 mg/kg)组。每天给药两次,连续7周。测定大鼠血压,胸主动脉舒张功能,血清一氧化氮(NO)、内皮素-1(ET-1)含量、血管内皮型一氧化氮合成酶(eNOS)基因的表达。结果霉茶总黄酮能降低SHRs血压;升高血清中NO含量,降低ET-1含量;上调组织中eNOS基因。结论霉茶总黄酮能改善SHRs血管舒张功能,其机制与上调胸主动脉eNOS基因表达水平,增加NO的合成有关。
Objective To study the effects of flavonoids from Myrsinaceae on the function of thoracic aorta in primary hypertensive rats (SHRs) and its mechanism. Methods SHRs were randomly divided into model group with low total flavonoids (90 mg / kg), medium (180 mg / kg), high dose (360 mg / kg) and compound OBZ (50 mg / kg). Administered twice daily for 7 weeks. The blood pressure, thoracic aortic diastolic function, serum nitric oxide (NO), endothelin-1 (ET-1) and vascular endothelial nitric oxide synthase (eNOS) gene expression were determined. Results Total flavonoids from Myrica rubra reduced the blood pressure of SHRs, increased the content of NO in serum, decreased the content of ET-1 and upregulated the eNOS gene in the tissues. Conclusion Myricetha flavonoids can improve the vasodilation function of SHRs, and its mechanism is related to up-regulation of eNOS gene expression in thoracic aorta and increase of NO synthesis.
