目的:研究CD4+ CD25highFoxp3+调节性T细胞(Treg)在胃癌患者外周血单个核细胞(PBMC)的比例变化,探讨其在抗肿瘤免疫调节中的作用及临床意义。方法:采用流式细胞术检测37例胃癌患者治疗前后PBMC中CD4+ CD25+/CD4+、CD4+ CD25high/CD4+ T细胞比例,以及CD4+ CD25+及CD4+ CD25highT细胞中Foxp3+ T细胞的比例,并与30例正常人对照组的上述指标进行比较。结果:胃癌患者治疗前PBMC中CD4+ CD25+/CD4+、CD4+ CD25high/CD4+ T细胞比例以及CD4+ CD25+及CD4+ CD25highT细胞中Foxp3阳性表达细胞的水平较正常人对照组明显增高(P<0.05),治疗后上述指标较治疗前明显减低(P<0.05),虽仍高于正常对照组,但差异无显著性(P>0.05)。结论:胃癌患者外周血调节性T细胞比例增高,可能是其免疫功能异常的主要原因之一。
Objective: To study the proportion of CD4 + CD25highFoxp3 + regulatory T cells (Tregs) in peripheral blood mononuclear cells (PBMCs) in patients with gastric cancer and to explore its role in anti-tumor immune regulation and its clinical significance. Methods: The proportion of CD4 + CD25 + / CD4 +, CD4 + CD25high / CD4 + T cells in PBMC and the ratio of Foxp3 + T cells in CD4 + CD25 + and CD4 + CD25high T cells in 37 gastric cancer patients before and after treatment were detected by flow cytometry. The above indicators of the group are compared. Results: The percentage of CD4 + CD25 + / CD4 +, CD4 + CD25high / CD4 + T cells and Foxp3 positive cells in CD4 + CD25 + and CD4 + CD25high T cells in gastric cancer patients before treatment were significantly higher than those in normal controls (P <0.05) The indexes were significantly lower than those before treatment (P <0.05). Although the indexes were still higher than those of the normal control group, the difference was not significant (P> 0.05). Conclusion: The increased proportion of T regulatory cells in peripheral blood of patients with gastric cancer may be one of the major causes of abnormal immune function.
