目的探讨外周抗凝全血中miRNA的表达与维吾尔族肥胖人群的关系。方法按照亚太地区推荐对肥胖诊断的标准,选择维吾尔族肥胖与非肥胖组抗凝全血样本各5例,采用芯片筛选出差异表达的miRNA;后扩大样本量,测量身高、体重、腰围、臀围后,留取血样检测空腹血糖、血脂等指标,采用实时PCR验证差异表达的miRNA。结果芯片结果显示,与非肥胖组相比维吾尔族肥胖组抗凝全血中miRNA130a的表达量下降〔(1.14±0.32)vs(0.28±0.13),P<0.05〕;维吾尔族肥胖组体重指数、腰围、臀围以及收缩压均大于非肥胖组,并且差异有统计学意义(P﹤0.05);实时PCR验证结果显示,与非肥胖组相比维吾尔族肥胖组抗凝全血中miRNA130a表达量降低〔(4.84±1.18)vs(2.03±0.59),P<0.05〕;相关分析结果在维吾尔族抗凝全血中miRNA130a的表达量与空腹血糖值、总胆固醇值呈负相关(P<0.05),而与高密度脂蛋白胆固醇呈正相关(P<0.05)。结论抗凝全血中miRNA130a的表达量在维吾尔族肥胖人群中显著降低,提示miRNA130a表达量降低可能与维吾尔族BMI有关。
Objective To investigate the relationship between miRNA expression in peripheral anticoagulant whole blood and Uygur obese population. Methods According to the recommended diagnostic criteria for obesity in the Asia-Pacific region, five samples of anticoagulated whole blood samples of Uygur obesity and non-obese patients were selected and differentially expressed miRNAs were screened by chip. The samples were then expanded to measure the height, weight, waist circumference, buttocks Surrounding, blood samples were taken for the detection of fasting blood glucose, blood lipids and other indicators, using real-time PCR to verify differentially expressed miRNA. Results The results showed that compared with non-obese group, the expression of miRNA130a in the whole blood of Uygur obesity group decreased (1.14 ± 0.32 vs 0.28 ± 0.13, P <0.05), and the body mass index Waist circumference, hip circumference and systolic blood pressure were significantly higher than those in non-obese group (P <0.05). Real-time PCR results showed that the expression of miRNA130a in Uighur obese group was lower than that in non-obese group (4.84 ± 1.18 vs 2.03 ± 0.59, P <0.05). The correlation analysis showed that the expression level of miRNA130a was negatively correlated with fasting blood glucose and total cholesterol in Uighur anticoagulant whole blood (P <0.05) But positively correlated with high density lipoprotein cholesterol (P <0.05). Conclusions The expression of miRNA130a in anticoagulated whole blood is significantly decreased in Uighur obese population, suggesting that the decrease of miRNA130a expression may be related to the Uygur BMI.
