血浆中脑啡肽(EK)含量极微且迅速酶解,测定很困难。本文报告了人血浆LEK样物质放射免疫测定值为127.9+5.4pg/ml(m±S. E. M,n=61)。作者用TG-ECDI-LEK背部皮内多点法免疫家兔,得高灵敏度、高特异性的抗血清;迅速酸化血浆防止EK酶介;XAD-2树脂吸附EK和水合甲醇洗脱提取。本法已通过多种方法学考验。作者观察到胰岛素低血糖试验和踏车运动激发血LEK升高;嗜铬细胞瘤患者血LEK增高,提示LEK可能来源于肾上腺髓质。本法为研究循环中EK变化提供了条件。
Plasma enkephalin (EK) content is minimal and rapidly digested, the determination is difficult. This paper reports the radioimmunoassay of human plasma LEK-like substance to 127.9 + 5.4 pg / ml (m ± S.E.M, n = 61). The author used TG-ECDI-LEK intradermal multipoint intradermal immunization of rabbits to obtain high sensitivity, high specificity antiserum; rapid acidification of the plasma to prevent EK enzyme mediated; XAD-2 resin adsorption EK and hydrated methanol elution extraction. This Law has passed a number of methodological tests. The authors observed elevated insulin levels in the hypoglycemia test and treadmill exercise, and elevated LEK in patients with pheochromocytoma, suggesting that LEK may be derived from the adrenal medulla. This Law provides the conditions for studying changes in EK in circulation.
