目的:探讨神经母细胞瘤荷瘤SD幼鼠成瘤规律及其原因分析。方法:以3~4周龄SD幼鼠为研究对象,随机分组为对照组(10只)和实验组(45只根据肿瘤生长情况,又再分成瘤组和非成瘤组)。每2 d观察并且测量各组生长指标,第20天处死动物后,计算各组胸腺、脾脏脏器指数;计算外周血淋巴细胞亚群CD3~+、CD4~+、CD8~+细胞数量变化;检测各组胸腺、脾脏组织中增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、Caspase-3蛋白的表达。结果:145只实验组SD幼鼠有13只成瘤,成瘤率为31.1%。2胸腺脏器指数:成瘤组>对照组(P<0.01),非成瘤组>对照组(P<0.01),成瘤组与非成瘤组无统计学差异(P>0.05);脾脏脏器指数在各组间无统计学差异(P>0.05)。3非成瘤组中CD3~+、CD8~+细胞数量明显高于成瘤组(P<0.01),非成瘤组与对照组、成瘤组与对照组中CD3~+、CD8~+细胞数量无统计学差异(P>0.05);CD4~+在3组中无统计学差异(P>0.05)。4胸腺组织中PCNA、Caspase-3蛋白表达:成瘤组及非成瘤组胸腺PCNA表达较对照组增加(P<0.05),Caspase-3表达较对照组降低(P<0.05),其中非成瘤组中PCNA较成瘤组明显增加(P<0.05),Caspase-3表达明显降低(P<0.05);在脾脏内PCNA、Caspase-3蛋白在各组中表达无统计学差异(P>0.05)。结论:1SD幼鼠接种人神经母细胞瘤细胞后,其生长规律出现潜伏期、快速生长期、平台期、消退期的变化;2成瘤率及成瘤规律与幼鼠免疫器官反应有关系。
OBJECTIVE: To investigate the tumorigenesis of neuroblastoma SD mice and its causes. Methods: The 3 to 4-week-old SD rats were randomly divided into control group (n = 10) and experimental group (n = 45), which were subdivided into tumor group and non-tumor group according to tumor growth. The indexes of growth of each group were observed and measured every 2 days. After the animals were sacrificed on the 20th day, the indices of thymus and spleen of each group were calculated. The numbers of CD3 +, CD4 +, CD8 + cells in the peripheral blood lymphocyte subsets were calculated. The expression of proliferating cell nuclear antigen (PCNA) and Caspase-3 protein in thymus and spleen of each group were detected. There was no significant difference of CD4 ~ + in the three groups (P> 0.05). 4 The expression of PCNA and Caspase-3 protein in thymus: Compared with control group, the expression of PCNA and thymus in non-tumorigenic group increased (P <0.05), Caspase-3 expression decreased (P <0.05) The expression of PCNA and Caspase-3 in the tumor group was significantly higher than that in the tumorigenesis group (P <0.05), and Caspase-3 expression was significantly decreased (P <0.05) ). CONCLUSIONS: The growth of 1SD rat showed changes in latency, rapid growth phase, plateau phase and regression phase after inoculation of human neuroblastoma cells; 2 The rate of tumor formation and tumor formation were related to the immune organ response in young rats.
