目的 :探讨bcl 2反义寡核苷酸诱导HL 6 0细胞的凋亡。方法 :将人工合成的bcl 2反义寡聚脱氧核苷酸片段与HL 6 0细胞共培养 ,在作用的第 0天、3天、5天通过免疫组化法 (APAAP法 )检测细胞内bcl 2蛋白表达水平的变化 ,通过流式细胞仪分析细胞凋亡的情况。结果 :bcl 2反义寡核苷酸可下调HL 6 0细胞内bcl 2的蛋白表达水平 ,其作用与其浓度和时间呈正相关 ,同时可诱导细胞凋亡 ,而bcl 2正义寡核苷酸及对照无此作用。结论 :bcl 2反义寡核苷酸诱导HL 6 0细胞凋亡的主要机制之一是下调bcl 2蛋白表达水平
Objective: To investigate the apoptosis of HL 60 cells induced by bcl 2 antisense oligonucleotides. METHODS: The synthetic bcl 2 antisense oligodeoxynucleotide fragment was co-cultured with HL 60 cells. The intracellular bcl was detected by immunohistochemistry (APAAP method) on the 0th, 3rd, and 5th day of action. 2 Changes in protein expression levels were analyzed by flow cytometry for apoptosis. RESULTS: bcl 2 antisense oligonucleotides could down-regulate bcl 2 protein expression in HL 60 cells. Its effect was positively correlated with its concentration and time. At the same time, it could induce apoptosis, whereas bcl 2 sense oligonucleotide and control No such effect. Conclusion: One of the main mechanisms of apoptosis of HL 60 cells induced by bcl 2 antisense oligonucleotides is down-regulation of bcl 2 protein expression level.
