目的探讨氨溴索对在体肺缺血-再灌注(I/R)损伤的保护作用。方法夹闭大鼠左肺门1.5 h,再灌注2 h,建立在体肺缺血-再灌注模型。将24只SD大鼠随机分成对照组(control组),缺血-再灌注组(I/R组)和氨溴索组(AMB组),AMB组缺血前30 min给予腹腔注射氨溴索25 mg/kg,再灌注前5 min颈静脉注射氨溴索25 mg/kg。再灌注2 h后采颈动脉血检测血气、白细胞介素-1β(IL-1β)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)。摘取左肺测湿干比、超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量、髓过氧化物酶(MPO)活力并行病理学检查。统计结果行两样本t检验。结果再灌注2 h后,动脉血氧分压和二氧化碳分压三组间差异无统计学意义;I/R组肺湿干比、MPO活力(U/g)、MDA含量(nmol/mg)分别为(5.3±0.5)、(1.30±0.26)和(0.66±0.16),显著高于control组,AMB组可使上述指标降至(4.8±0.3)、(0.93±0.40)和(0.51±0.07),同时使SOD(U/mgprot)由I/R组的(39.3±3.0)升高至(49.9±7.3)。I/R组血清IL-1β(pg/ml)、IL-8(pg/ml)、TNF-α(pg/ml)含量分别为(22.08±3.85)、(21.92±5.56)、(30.50±3.77)显著高于control组,AMB组可使上述指标降至(17.11±2.22)、(15.07+3.56)和(22.97±3.22)。结论氨溴索对肺缺血-再灌注损伤具有保护作用,可能与其抗氧化和抑制中性粒细胞和炎症因子分泌有关。
Objective To investigate the protective effect of ambroxol on lung injury induced by ischemia / reperfusion (I / R). Methods The left hilar of rats was occluded for 1.5 h and then reperfused for 2 h. The model of lung ischemia-reperfusion was established. 24 SD rats were randomly divided into control group, ischemia-reperfusion group (I / R group) and ambroxol group (AMB group). Conclusion Ambroxol has a protective effect on lung ischemia-reperfusion injury, which may be related to its antioxidation and inhibition of the secretion of neutrophils and inflammatory cytokines.
