目的:观察中药复方含药血清对模拟失重条件下体外培养成骨细胞的影响,探讨该方对抗骨丢失的部分作用机理。方法:分离乳鼠颅骨成骨细胞,体外培养后,随机分为5%,10%,20%浓度的空白血清对照组和5%,10%,20%空白血清回转组和5%,10%,20%含药血清回转组,连续回转48h模拟失重,观察该方对大鼠成骨细胞增殖、I型胶原合成、碱性磷酸酶(ALP)和骨钙素(BGP)生成的影响。结果:5%和10%空白血清回转组成骨细胞增殖比正常重力组明显降低(P<0.05);与10%空白血清回转模拟失重组相比,10%含药血清回转组可显著促进细胞增殖(P<0.05)。正常重力组与回转模拟失重组Ⅰ型胶原含量比较差异无统计学意义(P>0.05);与5%和10%空白血清回转模拟失重组相比,5%和10%含药血清回转组均能明显促进成骨细胞Ⅰ型胶原合成(P<0.05)。与5%正常重力组相比,5%空白血清回转组成骨细胞ALP活性明显降低(P<0.05);与5%和20%空白血清回转模拟失重组相比,5%和20%含药血清回转组能明显提高成骨细胞ALP活性(P<0.05)。与10%、20%空白血清组相比,10%、20%空白血清回转组成骨细胞BGP含量明显降低(P<0.05);而中药复方含药血清对其模拟失重成骨细胞BGP活性没有显著影响(P>0.05)。结论:该方能改善模拟失重条件下成骨细胞增殖,分化成熟,增加胶原蛋白合成,从而实现对抗失重骨丢失的效果。
OBJECTIVE: To observe the effects of Chinese medicine compound-containing serum on osteoblasts cultured in vitro under simulated weightlessness and to explore the mechanism of action of this compound on bone loss. Methods: Osteoblasts of suckling mice were isolated and cultured in vitro. The cells were randomly divided into 5%, 10% and 20% blank serum control groups and 5%, 10% and 20% , 20% serum-containing swirling group. The rats were continuously rotated for 48 hours to simulate weightlessness. The effects of the prescription on the proliferation of osteoblasts, synthesis of type I collagen, alkaline phosphatase (ALP) and osteocalcin (BGP) were observed. Results: Compared with the normal gravity group, the proliferation of osteoblasts in 5% and 10% blank serum swirling groups was significantly decreased (P <0.05). Compared with the 10% blank swirling simulated weightlessness group, 10% serum containing swollen groups could significantly promote cell proliferation (P <0.05). There was no significant difference in the content of type I collagen between normal gravity group and swirling simulated weightlessness group (P> 0.05). Compared with 5% and 10% blank serum swirling simulated weightlessness group, 5% and 10% Can significantly promote osteoblast type Ⅰ collagen synthesis (P <0.05). The ALP activity of osteoblasts in 5% blank serum swirling group was significantly lower than that in 5% and 20% blank serum swirling groups (P <0.05). Compared with 5% and 20% Swirling group can significantly improve osteoblast ALP activity (P <0.05). Compared with 10%, 20% blank serum group, the BGP content of osteoblasts in 10%, 20% blank serum group decreased significantly (P <0.05) Influence (P> 0.05). Conclusion: This recipe can improve the proliferation, differentiation and maturation of osteoblasts under simulated weightlessness and increase collagen synthesis, so as to achieve the effect of losing weightless bone.
