目的 :研究在光照下姜黄素诱导胃癌细胞凋亡的效应 ,探讨其作为光动力学治疗肿瘤新型光敏剂的可能性。方法 :体外培养的胃癌MGC80 3细胞加入姜黄素后光照处理 ;以台盼蓝拒染法计数细胞存活率 ,Giem sa染色观察形态变化并计数及统计凋亡指数 ,DNA琼脂糖电泳观察DNA片段化 ,流式细胞术检测细胞周期变化。结果 :光敏化姜黄素处理后 ,细胞存活率随药物浓度及处理时间依赖性下降 ,并呈现典型的凋亡细胞形态 ;5 μmol/L姜黄素光照处理细胞 2 4小时 ,即呈现典型凋亡梯形条带 ;经 2 5 μmol/L及 5 μmol/L光敏化姜黄素处理 ,细胞周期主要阻滞于G2 /M期 ,提示细胞的G2 /M期阻滞与光敏化姜黄素诱导MGC 80 3细胞凋亡有关。结论 :光敏化姜黄素具有显著诱导MGC 80 3细胞凋亡的作用 ,可望进一步开发成光动力学治疗肿瘤的新型光敏剂。
Objective: To study the effect of curcumin on gastric cancer cell apoptosis induced by light irradiation, and to explore its possibility as a novel photosensitizer for photodynamic therapy of tumors. METHODS: In vitro cultured gastric cancer MGC80 3 cells were treated with curcumin followed by light treatment. The viability of the cells was counted by trypan blue exclusion method. Morphological changes were observed by Giem sa staining. Apoptosis index was counted and counted. DNA fragmentation was observed by DNA agarose electrophoresis. Flow cytometry was used to detect cell cycle changes. RESULTS: After treated with curcumin, cell viability decreased with drug concentration and treatment time, and showed a typical morphology of apoptotic cells; 5 μmol/L curcumin treated with light for 24 hours showed typical apoptotic trapezoids. The band; treated with curcumin at a concentration of 25 μmol/L and 5 μmol/L, the cell cycle was mainly arrested at G2 /M phase, suggesting that G2 /M phase arrest and photocuring curcumin induced MGC 80 3 cells. Apoptosis related. Conclusion : Photosensitized curcumin can induce the apoptosis of MGC 80 3 cells significantly. It is hopeful to develop a novel photosensitizer for the photodynamic therapy of tumors.
