目的观察新风胶囊不同剂量组对佐剂关节炎(adjuvant arthritis,AA)大鼠肺功能、Th1/Th2细胞、调节性T细胞(Treg)的影响。方法将8 4只大鼠随机分为正常对照(NC)组、模型(M)组、甲氨喋呤(MTX)组、雷公藤多苷片(TGT)组和新风胶囊低剂量(XFC-L)组、新风胶囊中剂量(XFC-M)组、新风胶囊高剂量(XFC-H)组,每组12只,分别向除NC组外的其余动物右后足跖皮内注射弗氏完全佐剂0.1 m L致炎,致炎后第19 d开始给药。NC组及M组给予生理盐水,其余5组分别给予MTX、TGT、XFC。观察各组大鼠各指标的变化。结果 (1)与NC组比较,M组大鼠足跖肿胀度(E)、关节炎指数(AI)、肺系数(LI)、1秒内平均呼气流量(FEV1/FVC%)、肺泡炎积分、TNF-α、Th1/Th2细胞、TGF-β_1表达明显升高(P<0.01);用力肺活量(FVC)、25%肺活量的最大呼气流量(FEF25)、50%肺活量的最大呼气流量(FEF50)、75%肺活量的最大呼气流量(FEF75)、最大呼气中期流量(MMF)、用力最大呼气流量(PEF)、CD4~+Treg、CD4~+CD25~+Treg、IL-10、Foxp3表达明显降低(P<0.01)。(2)与M组比较,各治疗组大鼠体重、FVC、FEF25、FEF50、FEF75、MMF、PEF、IL-10及Treg、Foxp3的表达升高;E、AI、LI、FEV1/FVC%及TNF-α、Th1/Th2细胞、TGF-β_1表达降低(P<0.05,P<0.01)。(3)与XFC-M组比较,其他各治疗组LI、肺泡炎积分及TNF-α、Th1/Th2细胞、TGF-β_1升高;FVC、FEF25、FEF50、FEF75及IL-10、CD4~+Treg、CD4~+CD25~+Treg、Foxp3降低(P<0.05,P<0.01)。结论 AA大鼠在关节局部肿胀同时肺功能降低;新风胶囊能明显改善AA大鼠足跖肿胀度、关节炎指数和提高肺功能,其机制是通过促进IL-10、CD4~+Treg、CD4~+CD25~+Treg及Foxp3的表达、抑制TNF-α、Th1/Th2细胞及TGF-β_1的表达,降低炎症反应和免疫复合物对组织器官的损伤,改善关节和肺部症状。
Objective To observe the effects of Xinfeng Capsule on pulmonary function, Th1 / Th2 cells and Tregs in adjuvant arthritis (AA) rats. Methods Eighty four rats were randomly divided into normal control (NC) group, model group (M), methotrexate (MTX) group, tripterygium glycosides tablets (TGT) group and Xinfeng capsule low dose (XFC-M) group and XFC-H group (12 rats in each group). The animals in the other groups except the NC group were injected intradermally with Freund’s complete Agent 0.1 m L inflammation, the first 19 d after the start of inflammation. Rats in NC group and M group were given physiological saline, and the other five groups were given MTX, TGT and XFC respectively. The changes of each index in each group were observed. Results (1) Compared with NC group, the degree of paw edema (T), arthritis index (AI), pulmonary coefficient (LI), mean expiratory flow rate (FEV1 / FVC% (FVC), maximum expiratory flow (FEF25) of 25% vital capacity, maximum expiratory flow of 50% vital capacity FEF50, FEF75, MMF, PEF, CD4 ~ + Treg, CD4 ~ + CD25 ~ + Treg, IL-10 , Foxp3 expression was significantly lower (P <0.01). (2) Compared with M group, the expression of FVC, FEF25, FEF50, FEF75, MMF, PEF, IL-10, Treg and Foxp3 were increased in all treatment groups; TNF-α, Th1 / Th2 cells and TGF-β1 expression (P <0.05, P <0.01). (3) Compared with XFC-M group, LI, alveolitis score and TNF-α, Th1 / Th2 cells and TGF-β1 increased in other treatment groups; FVC, FEF25, FEF50, FEF75 and IL- Treg, CD4 ~ + CD25 ~ + Treg, Foxp3 decreased (P <0.05, P <0.01). Conclusions AA rats have local swelling in the joints with reduced lung function. Xinfeng Capsule can significantly improve the paw edema, arthritis index and pulmonary function in AA rats through promoting the expression of IL-10, CD4 ~ + Treg, CD4 ~ + CD25 ~ + Treg and Foxp3 expression, inhibit the expression of TNF-α, Th1 / Th2 cells and TGF-β_1, reduce the damage of tissues and organs by inflammatory reaction and immune complex, and improve the symptoms of joint and lung.
